Synapses are complex, highly specialized biological structures. Therefore, even slight functional disturbance can lead to disruptive brain disorders. Dysfunction of these structures is associated with a variety of aging-related neurodegenerative conditions, such as Alzheimer's and Parkinson's disease. Most synaptopathies compromise complex signaling pathways by progressively affecting the synaptic protein repertoire. Currently, the treatment of most of these pathologies focuses on the mitigation of symptoms; in consequence, further efforts need to be made to elucidate the common molecular culprit and cellular mechanisms underlying the cause of these severe pathologies to serve as therapeutic targets for clinical applications. In this Research Topic, we aim to discuss synaptic dysfunction as a causal factor for aging-related neurodegenerative diseases. Despite the recent technological advances that allow identifying the individual synaptic components affected in these synaptic pathologies, we know little about how they impact the synaptic function. To promote the discussion around the molecular and functional connection, we primarily welcome original research, review, and perspective articles emphasizing the relationship between synaptic function and the proteins or mechanisms affected.

The neuronal ceroid lipofuscinoses are an extremely rare group of inherited neurodegenerative diseases that primarily affect children. Core symptoms of these conditions typically include epilepsy, cognitive decline and visual failure. These diseases are so rare that professionals who come into contact with them need a consultative reference work that enables them to become expert, or identify who to contact for more details. Fully updated and revised, this second edition continues to be the definitive volume on this devastating group of disorders. Written by an international collection of authorities in the field, it provides invaluable advice on their diagnosis, patient care, and new treatments that are available. This new edition of the definitive reference text on the neuronal ceroid lipofuscinoses will prove useful for clinicians, family physicians, research scientists, diagnostic laboratories, families affected by the disease as well as by workers in industry planning translational research.

Intracellular cell signaling is a well understood process. However, extracellular signals such as hormones, adipokines, cytokines and neurotransmitters are just as important but have been largely ignored in other works. Aimed at medical professionals and pharmaceutical specialists, this book integrates extracellular and intracellular signalling processes and offers a fresh perspective on new drug targets.

The neurobiology and genetic section deals with epigenetics and intracellular pathways associated with etiology. The development and behaviour section deals with proprioceptive profiles and joint attention in autism. The final section focuses on interventions including mindfulness, animal assisted activity, social/cultural perspective on autism intervention and physical activity. The book is relevant to all professionals and researchers working with persons with autism, including psychiatrists/psychologists, speech and language therapists, occupational therapists, teachers, nurses and care workers.

It has been known for half a century that neurotransmitters are released in preformed quanta, that the quanta represent transmitter-storing vesicles, and that release occurs by exocytosis. The focus of this book is twofold. In the first part, the molecular events of exocytosis are analysed. In the second part of the book, the presynaptic receptors for endogenous chemical signals are presented that make neurotransmitter release a highly regulated process.

Synaptic Function written by Neurosciences Institute (New York, N.Y.) and has been published by Wiley-Interscience this book supported file pdf, txt, epub, kindle and other format this book has been release on 1987 with Medical categories.

This book consists of five sections. The first section details methods for analyzing both presynaptic and postsynaptic function and emphasizes the molecular aspects of synapses. It describes ongoing studies of neurotransmitter release, voltage- sensitive ion channels, and electronic transmission at gap junctions. The second section focuses on the growing menagerie of neurotransmitters: their catagorization into chemical families, their relation to ion channels, their modulation by second messenger systems and their role in pharmacologic action. The third section considers the important relationship of transmitter diversity and synaptic types to the behavior of actual cellular networks. All of the studies described in these sections point to the necessity of considering interactions between anatomy, chemistry, physiology and pharmacology if synaptic function is to be understood at any one of these levels of analysis.

With the unprecedented identification of new mutation mechanisms in neurodegenerative diseases and the emergence of common mechanisms among diseases that were once considered unrelated, neurobiologists are poised for the development of new therapies based on high throughput screenings and a better understanding of the molecular and cellular mechanisms leading to neurodegeneration. In Molecular Mechanisms of Neurodegenerative Diseases, Marie-Francoise Chesselet, MD, PhD, and a panel of leading researchers and neurologists from industry and academia critically review the most recent advances from different yet complementary points of view. Focusing on Alzheimer's, Parkinson's, and CAG triplet repeat diseases, the authors show how studies of cellular and genetically engineered animal models have enhanced our understanding of the molecular mechanisms of neurodegenerative diseases and may lead to the development of new therapeutics. Topics include the role of Ab toxicity, glial cells, and inflammation in Alzheimer's disease; the formation of abnormal protein fragments across several diseases, the impact of dopamine and mitochondrial dysfunction on neurodegeneration; and the potential of genetics to identify the molecular mechanisms of neurodegenerative diseases. Authoritative and insightful, Molecular Mechanisms of Neurodegenerative Diseases synthesizes the novel ideas and concepts now emerging to create a fresh understanding of neurodegenerative disorders, one that promises to lead to powerful new therapies that prevent, delay the onset, slow the progression, or even cure these cruel diseases.
