The immune system is a complex network in which different cell types and soluble factors interact to efficiently eliminate various kinds of microorganisms as well as aberrant cell clones. The roots of immunologic investigations reach far into the past. In 430 BC, Thucydides reported that survivors of the plague did not present a second time with similar symptoms. The first report of a successful immu- therapy was made by Edward Jenner in 1798 who found a protective effect of cowpox vaccination against human pox. Since then, much knowledge has been accumulated; today, investigations of the molecular mechanisms of immune regulation are of central research interest. The novel insights into gene polymorphisms and gene regulation gathered from this work has improved our knowledge of individual immune reactions and risk factors in overcoming infections. Strategies to use the immune system for cancer treatment have been propelled by the discovery of divergent immunoregulatory cytokines and the introduction of new gene therapy strategies to modify immune responses. Recently, the discovery of various dendritic cells has focused attention on these cell types as central elements of the immune response and to the possibility of dendritic cell expansion, maturation, and consecutive stimulation with immuno- active tumor-specific peptides. Similarly, methods for ex vivo expansion of various stem cell-derived cell types have led to an improved therapeutic management of various benign and malignant diseases.

Dendritic cells (DCs) play a critical role in immune system, as they are necessary both for innate and adaptive immunity. According to their function, dendritic cells can be classified in immune tolerogenic or inflammatory DCs. DCs have been shown to regulate T cell-mediated immune responses and lead to immune tolerance and autoimmunity. For example, immune-tolerogenic DCs facilitate the development of regulatory T cells and inhibit T helper 17-mediated autoimmunity in mice with experimental autoimmune encephalomyelitis. Moreover, inflammatory DCs activate CD8+ and CD4+ T cells and elicit T cell-mediated inflammatory immune responses in vivo. However, the molecular and cellular mechanisms underlying DC-mediated immune tolerance and autoimmunity are still obscure.

Since programmed cell death was first described in insects in 1964 and apoptosis was described in 1972, rapid progress has been made in understanding the basic mechanisms and genes regulating programmed cell death and apoptosis. In addition, defects in various genes regulating programmed cell death have been delineated in several experimental models of human diseases. This volume surveys various aspects of these rapidly developing areas of research in programmed cell death/apoptosis. This volume should be of interest to basic immunologists and molecular biologists. The volume begins with a historical perspective of cell death. The remainder of the volume is divided into four different parts. Part I deals with the signaling pathways in apoptosis, including cell cycle control of apoptosis, role of ceramide in apoptosis, role of antibody signaling, and biochemical regulation of apoptosis.
