Neurovirology, the study of viral infection of the ner vous system, has evolved at the interface of three of the most rapidly unfolding fields of investigation-neurobiology, vi rology, and immunology. In all three, increasing knowledge about the molecular structure of surface receptors, how in tracellular messages are transmitted, and how diversity is regulated genetically is provided, along with the techniques of molecular biology. This promises to give us knowledge not only about the process of infection and the complex host and viral determinants of neuroinvasiveness and neurovirulence, but eventually it will provide the background from which to engineer vaccines and to devise novel therapeutic agents. Animal virology and molecular biology developed quite independently from different origins. Animal virology was originally the province of the pathologists, and by clinical observation and histological preparations, they tried to ex plain the incubation period, the pathways of virus spread, and the mechanisms of disease. Molecular virology grew out of biochemistry, particularly through studies of bacterio phage, with emphasis on the physical and chemical structure of viruses and the sequences of biochemical events during the replicative cycle in cells.

In the summer of 1984, both of us were working with Professor Yechiel Becker in the Laboratory for Molecular Virology at the Hebrew University-Hadassah Medical center in Jerusalem. During a discussion about the increasing number of specialized journals and monographs, Dr. Becker pointed out that none covered both the clinical and molecular aspects of neurotropic virus infections, and he urged us to develop such a book with the help of colleagues who were conducting highly-regarded research in their individual areas related to neurotropic viruses. The responses to our request were gratifying, and each contribution provided both a comprehensive clinical description of the neurologic disease produced by a specific virus and an up-to-date review of the current research in the pathogenesis of the disease, with particular attention given to molecular mechanisms. Most, but not all chapters were written by clinical neurologists who applied basic science strategies and methodologies to the question of how neurotropic viruses produce disease. other chapters were written by virologists known for their longstanding commitment and expertise in the analysis of the pathogenesis of neurotropic virus infections. The present volume contains the Proceedings of an EMBO Workshop organized in June 1983 by the Institute of Virology, Veterinary Faculty, State University of Utrecht, The Netherlands. Some 70 scientists from 11 countries followed the invitation to present and discuss their recent data on the structure, replication, genetics and pathogenesis of coronaviruses. It was the second international meeting on these viruses; the Workshop, which was held in Zeist near Utrecht followed the example of the Wuerzburg symposium of October 1980. At that time it became clear that coronaviruses are unique in many respects. Once a group of viruses that were defined merely on the basis of their characteristic peplomer morphology, Coronaviridae family members are known today - to be constructed from essentially three polypeptides - to use a "nested set" of 5-6 subgenomic mRNAs in the expression of their large, positive and single stranded RNA genome, - to generate these subgenomic RNAs through specific fusion of non contiguous sequences, - to mature by budding from intracellular membranes, - to cause persistent infection with neurological involvement and sometimes immunopathological conditons. These and many other findings have been established only very recently. The articles collected in this book reveal and/or further detail these findings. Since these Proceedings contain the combined scientific presentations of representatives from virtually all laboratories engaged in the field, they provide a fairly comprehensive review of the state of the art in corona virology.

Viruses are studied either because they cause significant human, animal or plant disease or because they are useful materials for probing basic phenomena in biology, chemistry, genetics and/or molecular biology. Arenaviruses are unusually interesting in that they occupy both categories. Arenaviruses cause several human diseases known primarily as the hemorrhagic fevers occurring in South and Latin America (Bolivia: Machupo, Argentine, Junin virus, and Brazil: Sabia virus) and in Africa (Lassa fever virus). Because such viruses produce profound disabilities and often kill the persons they infect, they are a source of health concern and economic hardship in the countries where they are prevalent. Further, they provide new problems for healthcare persons owing to the narrowing of the world as visitors from many countries travel increasingly to and from endemic areas and may incubate the infectious agent taking it from an endemic area into an area where the virus is not expected. Such cases are now being re corded with increasing frequency. In addition to these hemor rhagic fever viruses, the arenavirus lymphocytic choriomeningitis virus (LCMV) can infect humans worldwide, although the illness is most often less disabling and severe than those elicited by the other arenaviruses. Yet, LCMV is of greater concern to non arenavirologists and experimentalists using tissue culture or ani mals, etc. , because normal-appearing cultured cells or tissues from animals used for research may be persistently infected with LCMV without manifesting clinical disease or cytopathology and may transmit that infection to laboratory workers.

Elucidation of the mechanisms of pathogenesis underlying the diseases caused by viruses and bacteria has fascinated scientists for many years in two ways. Firstly, these pathogenic agents represent relatively sim ple biological systems for the study of basic biological processes such as replication, gene regulation, genetic variability and host-pathogen interactions. Secondly, process in this field is valuable in a practi cal sence, since it can help in the control of these diseases. The avail ability of new genetic and immunological techniques, especially recom binant DNA methods and monoclonal antibody technology, has provided powerful tools for unravelling the genetic, biochemical and immunologi cal basis of viral and microbial pathogenesis. Molecular cloning has allowed the isolation of single genes or groups of genes related to phenotypes which appear to be immunologically important for pathogene sis. The specific elimination of such genes from the complex genomes of the pathogens can now be achieved with similar genetic techniques. These genetic studies have provided additional information on the role played by specific phenotypic traits in pathogenesis, especially when combined with relevant animal model systems. Furthermore, the struc tural analysis of important virulence factors and surface antigens may allow the prediction of antigenic domains suitable for the development of new vaccines. The 38th Mosbacher Colloquium focuses on the molecular basis of viral and microbial pathogenesis. The virology part begins with the well studied plant viroids. The unusual structure of their genome, as well as knowledge about their replication and pathogenicity, are presented.
