During vertebrate hematopoiesis many specialized cell types are formed with vastly different functions such as B cells, T cells, granulocytes, macrophages, erythrocytes and megakaryocytes. To tightly control the enormous proliferative potential of developing blood cells, an intricately balanced signaling and transcription network has evolved that ensures that the different cell types are formed at the right time and in the right numbers. Intricate regulatory mechanisms ensure that blood cells function properly and have a determined life span. Moreover, in the adaptive immune system, long-lived memory cells have evolved that ensure that when pathogens have been seen once they will never cause a problem again. In this book we will therefore make a journey from asking how more primitive organisms use the epigenetic regulatory machinery to balance growth with differentiation control towards digging deep into what controls the function of specialized cells of the human immune system. We will first discover that flies make blood but exist without blood vessels, why fish make blood cells in the kidney and which precise genetic circuitries are required for these developmental pathways. We will then learn the regulatory principles that drive the differentiation of mature blood cells from stem cells and what controls their function in mammals. In the process, we will find out what unites hematopoietic stem cells and endothelial cells. Finally, we will shed light on the molecular mechanisms that either alter hematopoietic cell differentiation or lead to the development of cells with impaired function.

Epigenetics of the Immune System focuses on different aspects of epigenetics and immunology, providing readers with the fundamental mechanisms relating to epigenetics and the immune system. This book provides in-depth information on immune cells as a toolbox in deciphering systematically regulated mechanisms using "omics" and computational biology approaches. In addition, the book presents the translational importance of epigenetics and the immune system in our understanding of pathophysiology in diseases and its therapeutic applications. - Provides an overview of most important immune mechanisms, the current status of epigenetics, and how both of them are brought together - Presents key principles of immune mechanisms in epigenetics, presenting current findings and key principles - Features in-depth chapter contributions from a wide range of international researchers and specialists in immunology, translational medicine and epigenetics - Merges two very large areas, covering the unique interrelatedness of epigenetics and immunology

Immune Biology of Allogeneic Hematopoietic Stem Cell Transplantation: Models in Discovery and Translation, Second Edition once again provides clinical and scientific researchers with a deep understanding of the current research in this field and the implications for translational practice. By providing an overview of the immune biology of HSCT, an explanation of immune rejection, and detail on antigens and their role in HSCT success, this book embraces biologists and clinicians who need a broad view of the deeply complex processes involved. It then moves on to discuss the immunobiology mechanisms that influence graft-versus-host disease (GVHD), graft-versus-leukemia effect, and transplantation success. Using illustrative figures, highlighting key issues, describing recent successes, and discussing unanswered questions, this book sums up the current state of HSCT to enhance the prospects for the future. The second edition is fully revised and includes new chapters on microbiome, metabolism, kinase targets, micro-RNA and mRNA regulatory mechanisms, signaling pathways in GVHD, innate lymphoid system development, recovery and function in GVHD, genetically engineered T-cell therapies, immune system engagers for GVHD and graft-versus-tumor, and hematopoietic cell transplant for tolerance induction in solid organ grafts.

Although much is known with respect to blood cell formation and function, many new concepts in the areas of the regulation of hematopoietic stem cell commitment and the subsequent survival, proliferation, and differentiation of progenitors have been elucidated in the last five years. Our understanding of the microenvironment where stem cells reside and commit to distinct blood types (the niche) has grown significantly in recent years. Furthermore, blood cells have been used as the key model system to study microRNA function and the role of microRNAs in the transformation of normal cells into cancer cells. The current volume Molecular Basis of Hematopoiesis, edited by Amittha Wickrema & Barbara Kee, provides the most recent developments in the area in addition to a chapter on the utilization of basic science knowledge for the treatment of blood diseases. Each chapter in this book has been written and edited by faculty in major academic and research institutions around the world, who are pushing the frontiers of research in this important area.
