The Third International Symposium on Neurotransmitter Receptors was held in Hiroshima at a time when the entire field of neurotransmitter receptors in the brain is progressing at an unprecedented pace. The sym posium also marked my retirement as Professor and Chairman of the Third Department of Internal Medicine, Hiroshima University School of Medicine, and a new beginning as a Professor of the University of the Air. The symposium was remarkably successful, and there were enthusiastic responses from scientists allover the world, proving that the meeting was timely. Neurons in the nervous system organize into complex networks and their functions are precisely controlled. The most important means for neurons to communicate with each other is transmission through chemical synapses, where the release of neurotransmitters by the presynaptic nerve terminal of one neuron influences the function of a second neuron. Since the discovery of chemical neurotransmission by Otto Loewi in the 1920s, great progress has been made in our understanding of mol- ular mechanisms of neurotransmitter release. The last decade has seen an explosion of knowledge in this field. The aim of Molecular Mechanisms of Neurotransmitter Release is to provide up-to-date, in-depth coverage of essentially all major mole- lar mechanisms of neurotransmitter release. The contributors have made great efforts to write concisely but with sufficient background information, and to use figures/diagrams to present clearly key concepts or experiments. It is hoped that this book may serve as a learning tool for neuroscience students, a solid reference for neuroscientists, and a source of knowledge for people who have a general interest in neuroscience. Glutamate is the most pervasive neurotransmitter in the central nervous system (CNS). Despite this fact, no validated biological markers, or biomarkers, currently exist for measuring glutamate pathology in CNS disorders or injuries. Glutamate dysfunction has been associated with an extensive range of nervous system diseases and disorders. Problems with how the neurotransmitter glutamate functions in the brain have been linked to a wide variety of disorders, including schizophrenia, Alzheimer's, substance abuse, and traumatic brain injury. These conditions are widespread, affecting a large portion of the United States population, and remain difficult to treat. Efforts to understand, treat, and prevent glutamate-related disorders can be aided by the identification of valid biomarkers. The Institute of Medicine's Forum on Neuroscience and Nervous System Disorders held a workshop on June 21-22, 2010, to explore ways to accelerate the development, validation, and implementation of such biomarkers. Understanding the mechanisms that underlie brain activity and function remains one of the major frontiers of biology. All the processes of how we co-ordinate our movements, sense our surroundings, react to stimuli and learn and retain information rely on complicated networks of neurons thatcommunicate with each other and their targets. This fast and accurate intercellular signalling most occurs at synapses, specialized processes of neurons that release chemical signals, called neurotransmitters. Neurotransmitters: Frontiers in Molecular Biology will provide the reader withextensive background information on neurotransmitter release. It takes a multidisciplinary approach, but does not assume previous knowledge having basic introductions to most topics. Topics however are covered in enough detail to be of interest to experts in the field. Throughout, emphasis is placedon the rationale by which proteins are assigned specific functions rather than just providing facts about function. The first chapter provides an introduction to the basic features and properties of the synapse and is followed by a chapter detailing several important techniques used to elucidatevarious aspects of release. Chapters 3 describes many of the biochemical approaches used to identify proteins involved in neurotransmitter release and then chapters 4 and 5 focus on more specific aspects of synaptic transmission: the proteins that transport neurotransmitters and the role ofphosphlipids in the process. The next five chapters concentrate on approaches to unravel the function of many proteins in vivo by using toxins, giant squid axons, C. elegans, Drosophila, and mice. The final chapter summarizes current knowledge on endocytosis and recycling. Knowledge of themolecular mechanisms underlying neurotransmitter release has expanded tremendously over the last 10 years. Many of the proteins involved have been isolated, but their roles have yet to be determined. Brain aminergic pathways are organized in parallel and interacting systems, which support a range of functions, from homoeostatic regulations to cognitive, and motivational processes. Despite overlapping functional influences, dopamine, serotonin, noradrenaline and histamine systems provide different contributions to these processes. The histaminergic system, long ignored as a major regulator of the sleep-wake cycle, has now been fully acknowledged also as a major coordinator of attention, learning and memory, decision making. Although histaminergic neurons project widely to the whole brain, they are functionally heterogeneous, a feature which may provide the substrate for differential regulation, in a region-specific manner, of other neurotransmitter systems. Neurochemical preclinical studies have clearly shown that histamine interacts and modulates the release of neurotransmitters that are recognized as major modulators of cognitive processing and motivated behaviours. As a consequence, the histamine system has been proposed as a therapeutic target to treat sleep-wake disorders and cognitive dysfunctions that accompany neurodegenerative and neuroinflammatory pathologies. Last decades have witnessed an unexpected explosion of interest in brain histamine system, as new receptors have been discovered and selective ligands synthesised. Nevertheless, the complete picture of the histamine systems fine-tuning and its orchestration with other pathways remains rather elusive. The transmission of the nervous impulse is always from the dendritic branches and the cell body to the axon or functional process. Every neuron, then, possesses a receptor apparatus, the body and the dendritic prolongations, an apparatus of emission, the axon, and the apparatus of distribution, the terminal arborization of the nerve fibers. I designated the foregoing principle: the theory of dynamic polarization (Cajal 1923). Ever since the beautiful drawings from Golgi and Cajal, we have been familiar with the organisation of neurones into dendritic, somatic and axonal compartments. Cajal proposed that these cellular compartments were specialised, resulting in his concept of ^dynamic polarisation'. He considered dendrites to be passive elements that simply transferred information from inputs to the soma. Since the discovery that dendrites of many neural populations release neuroactive substances and in doing so, alter neuronal output, it is now apparent that this theory requires qualification. This book presents recent developments in the neurophysiology of dendritic release of several chemical classes of transmitters in a number of different areas of the mammalian central nervous system. Once released from a neuron, these substances can act as neurotransmitters and/or neuromodulators, to autoregulate the original neuron, its synaptic inputs, and adjacent cells or, by volume transmission, to affect distant cells. In some systems, dendritic transmitter release is part independent of secretion from axon terminal signifying a selective control of the dendritic compartment.
