New Trends in Genetic Risk Assessment is based on the Fifth International Round Table of the Rhône-Poulenc Santé Foundation, held in Nice 1987. The conference was an attempt to review the latest theories and mechanisms stipulated for the various aspects of genotoxicity; it was above all an open forum to discuss the new trends, the new tests, and the new battery of tests for assessing the genetic risk of chemicals and especially drugs. This volume is actually not a proceedings of the meeting but a monograph specially edited to report the reviews which were presented and the discussions which took place; it was designed to provide a better understanding of the knowledge obtained in the most recent years and to help in the practical choice of approaches or tests for the prediction of the various forms of genotoxicity. The main subjects of this review concern molecular analysis of mutagenesis, detection of DNA damage, gene mutation, clastogenesis, aneuploidy, and germ cells. Each theme is preceded by a short overview summarizing the state of the art and the contributions of each author. Finally, special attention was given to the personal views of some leading toxicologists as to the battery of tests presently available or recommended.

Much of organic chemistry is based on the ability of suitably structured chemicals to bind together through the formation of covalent bonds. Biochemistry is replete with exam ples of enzymatically catalyzed reactions in which normal body constituents can be linked through covalent bonds during the process of intermediary metabolism. The finding that xenobiotic chemicals that enter the body from the environment, are metabolized to highly reactive species, and then covalently react with cellular macromolecules to induce toxic and carcinogenic effects was an observation that spawned the research featured in the Fifth International Symposium on Biological Reactive Intermediates (BRI V). The group of investigators that became fascinated with this process and its signifi cance in terms of human health began their discussions in Turku, Finland (J 975), and continued them at Guildford, England (1980), College Park, Maryland (1985), Tucson, Arizona (1990), and Munich, Germany (1995). The first two editions of this book, published in 1979 and in 1986, were well re ceived by the scientific community. Translations into Italian, Japanese, and Rus sian suggest that this book was regarded useful in many parts of the world. Mean while, human genetics has seen dramatic developments, and the "molecular revo lution" has attracted thousands of scientists, including many molecular biologists, to this field. About 3700 human genes have already been mapped to chromosomal sites. Many such genes have been cloned, and the various mutations causing dis ease have been identified. Novel mutational mechanisms such as expanded trinu cleotide repeats have been discovered in conditions such as Huntington's disease and the fragile X syndrome of mental retardation. Gene action now can often be elucidated by studying the pathway from gene to phenotype following positional cloning rather than working in the opposite direction, as was customarily done be fore the tools of "new genetics" were available. In an increasing number of genetic diseases, the pathogenic mechanisms have been elucidated with positive conse quences for prevention and treatment. It therefore became necessary to rewrite al most completely major portions of this book. These developments are now making genetics arguably the leading basic science for medicine, as well as a recognized medical speciality. But all these changes do not mean that the entire framework of human genetics had to be reconstructed.

Genetically engineered (GE) crops were first introduced commercially in the 1990s. After two decades of production, some groups and individuals remain critical of the technology based on their concerns about possible adverse effects on human health, the environment, and ethical considerations. At the same time, others are concerned that the technology is not reaching its potential to improve human health and the environment because of stringent regulations and reduced public funding to develop products offering more benefits to society. While the debate about these and other questions related to the genetic engineering techniques of the first 20 years goes on, emerging genetic-engineering technologies are adding new complexities to the conversation. Genetically Engineered Crops builds on previous related Academies reports published between 1987 and 2010 by undertaking a retrospective examination of the purported positive and adverse effects of GE crops and to anticipate what emerging genetic-engineering technologies hold for the future. This report indicates where there are uncertainties about the economic, agronomic, health, safety, or other impacts of GE crops and food, and makes recommendations to fill gaps in safety assessments, increase regulatory clarity, and improve innovations in and access to GE technology.
