Phagocytosis—Past and Future is a collection of contributions by investigators who met in the Province of Messina, Sicily, in October 1980 to discuss the functions of phagocytic leukocytes in the host-parasite relationship. The topics discussed are largely in the areas of cell biology, cellular immunology, and biochemistry. They ranged from aspects of cellular movement and the ingestion process per se, to definition of substances that affect the function of phagocytic leukocytes and the nature of cidal mechanisms at the molecular level. This volume is organized into six sections encompassing 33 chapters and begins with an overview of Ilya Ilich Metchnikoff's life as well as his scientific contributions, particularly in the area of bacteriology and his research on mesodermal amoeboid cells. The next chapters focus on the mechanism of phagocytosis, with reference to aspects of cell membrane, cell movement, and ingestion. The reader is methodically introduced to the biochemical aspects of phagocytosis, from molecular modulations to energetics, oxygen radicals, and enzymes. The discussion then shifts to macrophages, their function in antigen recognition, receptor-mediated endocytosis in macrophage cell hybrids, and the role of phagocytosis in macrophage activation. The remaining chapters examine the cidal mechanisms of phagocytic leukocytes and aspects of inflammation. This book will be of interest to scientists and researchers in fields such as biology, biochemistry, immunology, pathology, pharmacology, and physiology.

Phagocytosis has been at the forefront of cell biology for more than a century. Initially, phagocytosis, which comes from Greek words meaning “devouring cells,” was discovered in the late 19th century by Ilya Metchnikoff, who was awarded, together with Paul Ehrlich, the Nobel Prize in Physiology and Medicine in 1908 “in recognition of their work on immunity.” At that time Metchnikoff had already identified a function for phagocytes not only in host defense but also as scavengers of degenerating host cells during metamorphosis of tadpoles, thus providing one of the first descriptions of apoptotic cell clearance by macrophages (Kaufmann 2008). Since then, much has been learned about phagocytosis, and the previous several decades have witnessed outstanding progress in understanding the functions and the molecular mechanisms of phagocytosis. Two main types of targets are cleared by phagocytosis: microbial pathogens and dying cells. Rapid recognition and clearance of dying cells by phagocytes plays a pivotal role in development, maintenance of tissue homeostasis, control of immune responses, and resolution of inflammation. Clearance of dying cells can be divided into several stages, including sensing, r- ognition, binding and signaling, internalization, and immunological responses. In this book, our contributors address these different stages of dead cell cle- ance and examine how impaired clearance of dying cells may lead to human d- eases. We have attempted to provide sufficient cross-referencing and indexing to enable the reader to easily locate the ideas elaborated in the different chapters.

This filed of study is growing rapidly after a somewhat slow start over recent decades. This collection of invited chapters attempts to reflect current research, and brings together cell biologists, microbiologists and immunologists with disparate interests. However, there is a certain unity, even repetition of key themes, hopefully like a symphony rather than a boring catalogue. It will be evident that editorial bias favors intracellular paratism and medically important organisms. The neutrophil is far more than a supporting player to the macrophage, and some attempt is made to remind the reader of some of its unique skills. To retain a manageable size, the emphasis is on relatively early events such as mutual recognition, cell entry, and response, rather than on longterm changes in gene expression by either host cell or pathogen. Viruses are excluded not because of lack of importance but because of somewhat different research approaches, although it is cytogenes, share common strategies in invasion and intercellular spread.

This comprehensive treatise on the reticuloendothelial system is a project jointly shared by individual members of the Reticuloendothelial (RE) Society and bio medical scientists in general who are interested in the intricate system of cells and molecular moieties derived from those cells which constitute the RES. It may now be more fashionable in some quarters to consider these cells as part of what is called the mononuclear phagocytic system or the lymphoreticular sys tem.
