This study was undertaken to identify and study novel linker histone variants in Arabidopsis thaliana. It was specifically focussed on determining tissue specificity, and possibly identifying an Arabidopsis cognate of a meiosis-specific, linker-histone-like protein called meiotin-1, previously characterized in Lilium longiflorum. Amino acid sequence similarity to the linker histone globular domain identified five candidate proteins. Two of those candidates (BT and F20) showed weak similarity to Meiotin-1, and were selected for further study; a third gene (BTBB) was also included, due to its close similarity to BT. Analysis of the three genes indicated that they were unlikely to encode meiotin-1 cognates. Semi-quantitative RT-PCR showed no meiosis-specificity in the expression of any of the genes, but BTBB did appear to be expressed in tissues undergoing active division. These novel genes probably encode chromatin associated proteins; BT and BTBB appear to be related to plant HMGA proteins, while the function of F20 remains elusive.
