The theses, lectures and solved problems within this book give an introduction to principles of mass spectrometry related to the analysis of biomolecules. The book focuses on MS and MS/MS analysis of peptides, oligosaccharides, lipids, glycoconjugates and polyphenols. The basic properties of modern MS ionization techniques and mass analyzers are presented, as well as appropriate MS methods for each class of biomolecules in modern biomedical scientific research. The biggest part of this book is focused on MS/MS analysis of biomolecules: how the MS and MS/MS data are generated, what are the basic strategies for their structural analysis, as well as how to interpret the respective MS and MS/MS data. This book also covers principles and examples of advanced MS/MS analyses, serving as a platform towards an intermediate level of understanding scientific research carried out in the field of structural MS analysis of biomolecules. It is the author's hope that the reader will be able to understand principles of MS analyses related to their scientific and/or analytical interest, as well as design/help to propose MS and MS/MS experiments, as well as interpret the obtained data. This book is primarily written for students with basic knowledge of general, organic and analytical chemistry, and who are interested in applying MS-based methodologies in biomedical research fields, as well as interested in pursuing a career in mass spectrometry. Therefore it is best suited for senior undergraduate students and first/second year Ph.D. students. However, since MS instrumentation is rapidly adopted on all levels of scientific as well as analytical laboratories, the book should prove useful to scientists -- as well as analysts -- interested in applying MS in their upgraded laboratories. The author hopes that the reader, after studying the content presented, will be able to read primary scientific literature containing MS data to learn more about specific topics of interest, as well as be more capable of choosing a MS-related career path. For senior scientists, this book can offer some basic insights related to choosing a post-doc laboratory, or simply easier and more fruitful cooperation with mass spectrometrists.

The first systematic summary of biophysical mass spectrometrytechniques Recent advances in mass spectrometry (MS) have pushed the frontiersof analytical chemistry into the biophysical laboratory. As aresult, the biophysical community's acceptance of MS-based methods, used to study protein higher-order structure and dynamics, hasaccelerated the expansion of biophysical MS. Despite this growing trend, until now no single text has presentedthe full array of MS-based experimental techniques and strategiesfor biophysics. Mass Spectrometry in Biophysics expertly closesthis gap in the literature. Covering the theoretical background and technical aspects of eachmethod, this much-needed reference offers an unparalleled overviewof the current state of biophysical MS. Mass Spectrometry inBiophysics begins with a helpful discussion of general biophysicalconcepts and MS-related techniques. Subsequent chaptersaddress: * Modern spectrometric hardware * High-order structure and dynamics as probed by various MS-basedmethods * Techniques used to study structure and behavior of non-nativeprotein states that become populated under denaturingconditions * Kinetic aspects of protein folding and enzyme catalysis * MS-based methods used to extract quantitative information onprotein-ligand interactions * Relation of MS-based techniques to other experimental tools * Biomolecular properties in the gas phase Fully referenced and containing a helpful appendix on the physicsof electrospray mass spectrometry, Mass Spectrometry in Biophysicsalso offers a compelling look at the current challenges facingbiomolecular MS and the potential applications that will likelyshape its future.

Many fundamental aspects of the methods used in mass spectrometry are here presented by outstanding scientists, with reference to very recent developments. The principles and applications of electrospray, ion spray and MALDI ionization technique are presented, together with optimised GC/MS interfacing systems and tools for quantitative analysis. A comprehensive treatment of modern instrumentation for mass analysis and detection is also included. The major part of the book deals with bioanalytical applications to peptides, proteins, oligonucleotides, polysaccharides, lipids and plant metabolites. Several papers are devoted to the evaluation of adduct formation between DNA and carcinogens. Environmental applications are also included, with examples of some specific cases. Fundamentals and applications are treated with the same degree of depth: the first two parts of the book therefore provide a basis for the understanding of the biomolecular applications section. Audience: Ideal for advanced graduate students of chemistry who have learned some basic mass spectrometry. Also useful for Ph.D. students in chemistry, biology and medicine. Of value to researchers in academic and industrial laboratories.

The developments in mass spectrometry over the past fifteen years have been impressive in their implications in bioanalytical chemistry. The achievements begin with the inventions of Cf-252 Plasma Desorption Mass Spectrometry by Macfarlane and Fourier Transform Mass Spectrometry by Comisarow and Marshall in the mid 1970s. The former showed the feasibility of producing large gas-phase ions from large biomolecules whereas the latter enhanced the capabilities for ion trapping especially in analytical mass spectrometry. A major achievement was the development by Barber of Fast Atom Bombardment (FAB) mass spectrometry, an advance that heralded a new era in biological mass spectrometry. Contemporary and routine instruments such as magnetic sectors and quadrupoles were rapidly adapted to F AB, and nearly the entire universe of small molecules became amenable to study by mass spectrometry. The introduction of FAB also paved the way for improvement of instrument capability. For example, the upper mass limit of magnet sector mass spectrometers was increased by nearly an order of magnitude by the instrument manufacturers. Furthermore, the technique of tandem mass spectrometry (MS/MS) was given new meaning because important structural information for biomolecules could now be produced for ions introduced by FAB into the tandem instrument. The evolution of MS/MS continues today with the development of ion traps, time-of-flight, and sector instruments equipped with array detection.
