Ageing is the main reason for most neurodegenerative diseases, including Alzheimer’s disease (AD) and Parkinson’s disease (PD). About 10% of individuals aged ≥65 years develop AD, which continuously increases with growing age. Medicinal plants have been used for age-related neurodegenerative disorders for a long time. These plants are rich in terms of various phytochemical constituents such as flavonoids, sterols, alkaloids, saponins, and terpenoids. Different scientific studies have been carried out on these medicinal herbs, e.g., anti-inflammatory, antioxidant, and neuro-regenerative activities that may be used in the treatment of AD and other age-related diseases. These medicinal plants, e.g., Curcuma longa, Bacopa monniera, Glycyrrhiza glabra, Commiphora whighitti, and Acorus calamus L., have anti-inflammatory properties that may decrease swelling of the brain tissue in AD. Despite this, there is currently no book available in the market that provides full information on the phytochemical and medicinal uses of the medicinal plants exclusively used for the management of neurodegenerative diseases in the elderly. This book aims to provide some important and evidence-based information on the plants used for neurodegenerative diseases in the elderly, especially AD and PD, in a planned manner so the academicians, researchers, and students working in the fields of medicinal plants, plant science, chemistry, biotechnology, pharmacognosy, pharmaceuticals, biochemistry, and many other interdisciplinary subjects may benefit. Epigenetics refers to changes in the expression of heritable genes or cellular phenotypes caused by regulating DNA methylation, histone modification, chromatin remodeling, and noncoding RNA expression, without altering the DNA sequence. It plays a critical role in the regulation of many physiological and pathological processes, such as inflammation, immunity, and tumor. In particular, many epigenetic modifiers have been found to affect innate cell development, infection, and inflammatory responses by regulating gene-specific expression. Epigenetic abnormalities often result in changes in gene expression and its function, which in turn promote the process of inflammation and even tumorigenesis. Autophagy is a highly conserved subcellular degradation process by which some damaged organelles, misfolded proteins, nucleic acids, and pathogenic microorganisms are digested to maintain homeostasis. Autophagy dysregulation has been implicated in multiple diseases, including inflammation, tumors, and neurodegenerative diseases. It is not only a cytoplasmic event, but also involved in the regulation of nuclear components, including histone modifications, microRNA, and transcription factors. Methyltransferase-mediated histone modifications (H3K9me2, H3R17me2, and H3K27me3), miRNAs, and DNA methylation catalyzed by DNA methyltransferase are the key nuclear factors involved in the regulation of autophagy. In addition, some autophagy-related genes (Atgs) promote or inhibit the autophagic process through epigenetic modifications, such as histone modifications and DNA methylation, affecting the occurrence and development of inflammatory diseases. Studying the role of epigenetic regulation of autophagy in inflammatory diseases may help identify novel drug targets and develop potential diagnostic and therapeutic approaches. Although the biological and clinical significance of epigenetic modification of autophagy has attracted more and more attention from researchers, this regulation is very complex and its role and mechanism in inflammatory diseases are still unclear.

Several types of brain injuries are causes of acquired temporal lobe epilepsy (TLE). The seizure-free "latent period" that often follows the brain injury is of unknown mechanistic significance but is commonly considered as the "epileptogenic" period characterized by gradual pathogenic processes leading to the onset of clinically detectable epilepsy. Acute convulsive status epilepticus (SE) is often associated with an adverse developmental outcome characterized by learning disabilities related to the cumulative effects of seizures and development of TLE. The symptomatic manifestations of TLE appear only after a widespread irreversible damage of entorhinal cortex, and hippocampus, the brain area most affected by this disease. These pathological features of TLE reduce the possibility of successful therapeutic approaches, often rendering the disease refractory. The difficult clinical management of chronic TLE and the limited success rate of surgical approaches, increase the incapacitating nature of this specific epileptic disorder. Prevention of TLE with an appropriate intervention after a known inciting event (in the case of acquired epilepsy) might represent the most ambitious goal in the clinical treatment of this epileptic disorder, but has been largely unsuccessful to this point. Clinical trials aimed at prevention of chronic epilepsy have often produced negative, disappointing results. However, in most cases, these studies ultimately evaluated the downstream clinical manifestations, failing to monitor early, specific molecular epileptogenic events. Therefore, elucidation of the underlying mechanisms of epileptogenesis, and their time course(s) are essential. The primary purpose of this topic is to collect scientific contributions providing novel insights in the cellular and molecular mechanisms of epileptogenesis as potential targets for innovative therapeutic approaches aimed at preventing the chronic epileptic disorder.
