Synthetic polymers make excellent specimens for light microscopy. Despite this, the use of the technique, at least in its advanced forms, is not so widespread as might be expected. Although reliable and relevant data are difficult to find and quantify, it seems that in other fields of materials science and technology there is a greater readiness to tum to the microscope in research, in industrial problem solving, or for quality assessment and control. It also seems that the reasons for the present situation are partly historical, partly the result of the structure of the plastics and rubber industries, and partly the education and training background of senior staff who tend to be chemistry or engineering based. In neither field does light microscopy feature strongly in the basic training. The primary aim of this book is to provide some insight into the range oflight microscopy techniques applicable to polymeric specimens, and to highlight typical applications to commercial polymers and polymer products. Where appropriate, the optical techniques involved are discussed in some detail. However, it has not been the intention to produce a light microscopy textbook dealing with the principles and design of the basic instrument. Many such texts are available, and selected examples are cited in the reference list at the end of most chapters.

This textbook provides an accessible and pedagogical explanation of the way microscopes magnify images and covers all techniques to date in transmitted and fluorescent light microscopy. The first section covers basic optics as it relates to microscopy. The second section describes all the major optical techniques of transmitted light microscopy, starting with brightfield, through darkfield, polarized light, phase contrast, differential interference contrast, and Hoffman modulation contrast microscopy. The final third of the book covers all the techniques of fluorescence microscopy. It begins with a simple explanation of fluorescence and is followed by Widefield epifluorescence, confocal, and 2-photon microscopy. This is followed by computational imaging including restoration ('deconvolution') microscopy, and all the present super-resolution techniques. The book concludes by discussing attainable resolution using transmitted light microscopy, including a thorough discussion of the Rayleigh, Sparrow, and other criteria of resolution, ending with a short discussion of the common pitfalls that result in decreased microscope resolution. The final chapter in the book is a short history of the microscope, beginning with the ancients, then discussing three seminal natural philosophers: Leeuwenhoek, Swammerdam, and Hooke. The remaining sections of the history chapter cover mechanical and optical advancements in the history of transmitted light microscopy. Finally, it includes a short history of fluorescence microscopy starting with Köhler and Rohr's first use of fluorescence microscopy in 1905, and ending with a description of the Sarastro Phoibos 1000, the first commercial confocal microscope.

Up to about twenty-five years ago, virtually the entire field of microscopy could be overseen and even practized by any active research worker. The rapid evolution which microscopy in its broadest sense has since undergone and which has contributed greatly to our insight in many fields of biological science and medicine has, however, lead to a progressive specialisation. Both experienced investigators in clinical and biological laboratories and post graduate students, confronted with a limited number of microscopic tech niques in their daily research work, have increasing difficulty in keeping (or obtaining) a general idea of the many time-honoured and new possibilities which microscopy has to offer. This book has been written with the aim of presenting general informa tion on light microscopic techniques, at a level somewhere in between booklets like those provided by microscope manufacturers (which are often too much focussed on the production program of a particular make) and very advanced treatises with a thorough mathematical treatment of all phenomena concerned. The physically oriented texts moreover often do not sufficiently take into account the practical situation in a medical or biolog ical laboratory; on the other hand, the value of really understanding what one is doing in using a microscopic technique is often underestimated. At tempt has been made, therefore, to present sufficient background informa tion necessary for a rational application of the different microscopical tech niques in their mutual relationship.
