Receptor-Mediated Binding and Internalization of Toxins and Hormones presents the proceedings of a conference conducted for the purpose of exploring relationships between hormones and toxins, specifically, receptor-mediated internalization of toxins and hormones and the role of this internalization in message transmission and biodegradatory processes. This book is divided into three sections, with the first focusing on model systems of internalization. It presents pictorial illustrations of the receptor-mediated endocytosis of low-density lipoprotein via coated regions of membrane and discusses the receptor for lysosomal enzymes on the surface of cultured fibroblasts. Conventional attempts to control cell behaviour and function are often marred by the toxicity of the drugs used, their premature waste or inactivation or by their inability to interact with or reach target sites efficiently. New trends in pharmacology empha size the development of methods for the optimization of drug action, for instance by the delivery of drugs, enzymes, hormones, antigens, genetic material, ets. through carrier systems selectively to re levant cellular and subcellular sites. A wide assortment of carriers was discussed in the first NATO Advanced Studies Institute (ASI) "Targeting of Drugs," the proceedings of which were published by Plenum Press in 1982 (eds., G. Gregoriadis, J. Senior and A. Trouet). This book, containing the proceedings of the 2nd NATO ASI "Receptor-Mediated Targeting of Drugs" held again at Cape Sounion, Greece during 20 June-l July 1983, deals with drug delivery through systems possessing ligands which can recognize and interact with receptors on the target's surface. This volume focuses exclusively on those endocytic processes that sequester proteins by a selective, receptor-mediated mechanism. In such an endocytic process, cell surface receptors specifically bind protein ligands and localize them to specialized invaginations of the plasma membrane. These regions are coated pits, so named because they are lined on the cytoplasmic face with an ordered array of the protein, clathrin. It is this 'coat' which provides their characteristic electron microscopic image. Subsequently, these regions pinch off to form coated vesicles which rapidly lose their 'coat' and then fuse with other organelles or the plasma membrane. The hallmarks of ,this process are the specific receptors, coated pits, coated vesicles and an ordered sequence of transit events leading to delivery to selected locations. Receptor recognition, specific disposition of the endocytosed ligand and the existence of recep tor-ligand complexes at highest density in coated pits define the process as selective and concentrative. This topic has received ever increasing attention during the past few years. The evolving mechanisms are especially exciting because they come at a time when the conventional views based on thermodynamic arguments suggest that proteins should not be able to cross into the cell. Receptor-mediated endocytosis, however, reconciles the view that biological membranes should be impervious to macromolecules with the evidence that certain mac romolecules do gain entrance into the cell. During the last few years this field has been stimulated by studies on the uptake and processing of low density lipoproteins (LDL) by cells.

The eleventh monograph and meeting of the Foundation on "Hormon al Control of the Hypothalamo-Pituitary-Gonadal Axis" was held in October 1983 at the Weizmann Institute of Science. This monograph honors the memory of Professor Hans R. Lindner, a productive and innovative scientist greatly respected and admired by his col leagues. When addressing the opening session I remarked that my impres sion of the Weizmann Institute was one or two large buildings hous ing the various departments. This was my first visit to Israel and I was overwhelmed by the beautiful semi-tropical gardens of the in stitute, in a setting of shrubs and trees, orange groves and flow ers. Sited among this seventy-five acres are over fifty buildings and residential areas for the staff and visiting scientists. I saw pictures of this area when Dr. Chaim Weizmann founded in 1934, the Daniel Sieff Research Institute, the forerunner of the Weizmann In stitute. The site was sand dunes without a blade of grass. That the desert shall bloom is illustrative of the progress made in Is rael. The topics of the monograph are grouped into twelve sections.
