This work describes the current knowledge of biochemical mechanisms regulating initiation of DNA replication in Escherichia coli, which focuses on the control of activity of the DnaA protein. Examples of direct linkages between DNA replication and other cellular processes are provided. In addition, similarities of the mechanisms of regulation of DNA replication operating in prokaryotic and eukaryotic cells are identified, and implications for understanding more complex processes, like carcinogenesis are suggested. Studies of recent years provided evidence that regulation of DNA replication in bacteria is more complex than previously anticipated. Multiple layers of control seem to ensure coordination of this process with the increase of cellular mass and the division cycle. Metabolic processes and membrane composition may serve as points where integration of genome replication with growth conditions occurs. It is also likely that coupling of DNA synthesis with cellular metabolism may involve interactions of replication proteins with other macromolecular complexes, responsible for various cellular processes. Thus, the exact set of factors participating in triggering the replication initiation may differ depending on growth conditions. Therefore, understanding the regulation of DNA duplication requires placing this process in the context of the current knowledge on bacterial metabolism, as well as cellular and chromosomal structure. Moreover, in both Escherichia coli and eukaryotic cells, replication initiator proteins were shown to play other roles in addition to driving the assembly of replication complexes, which constitutes another, yet not sufficiently understood, layer of coordinating DNA replication with the cell cycle.

In numerous conversations with our colleagues from India, it was suggested that we help to institute a series of symposia in India similar in nature to those that have been conducted by our Latin American colleagues for more than 10 years. We were fortunate to have with us in Oak Ridge Dr. Niyogi and Dr. Mitra from Indian universities. Their close ties with the Bose Institute in Calcutta and the resultant correspondence with the Institute Director, Dr. S. M. Sircar, provided the stimulus for organization of this first Indian symposium, which was held in Calcutta. Under the direction of Dr. Sircar, Dr. B. B. Biswas did an outstandingjob of organizing this conference. Financial support was arranged through Dr. R. R. Ronkin of the United States National Science Foundation, who smoothed the way for the use of PL 480 funds which were approved by the Indian Government for the organization and running of this most valuable symposium. The many Indian scientists who contributed papers and enthusiastically and vigorously entered into the discussions demonstrated the strength of modern science in India. The topic, Control of Transcription, is a timely one, and considerable activity in this area is going on all over the world. The success of this symposium speaks well for the future of these Indian conferences and workshops being planned for the next few years. Again, the worldwide "community of science" is clearly manifested by the close cooperation we have observed in this fruitful and successful symposium.

My aim in writing Gene Function has been to present an up-to-date picture ofthe molecular biology of Escherichia coli. I have not attempted a chronological description, believing that a mechanistic account is more useful for such a highly developed field. I have divided the book into four parts. Part I is a general introduction to bacterial systems, their genetic material, structure, composition and growth. It has seemed desirable to include herein a brief preview of the remaining text, to introduce the nomenclature and to help place subsequent chapters in perspective. The expression of genetic material and its perturbation through mutation is considered in Part II. Part III discusses how the transfer of prokaryotic genetic material can be mediated by plasmids and bacteriophages. It describes the DNA transactions involved (replication, recombination and repair) and ends with a description of the genetic and biochemical techniques employed in the study of gene organisation. Finally, Part IV considers the control of expression of bacterial, plasmid and phage genes. Key reviews are listed at the end of each chapter.
