Co-chaperones are important mediators of the outcome of chaperone assisted protein homeostasis, which is a dynamic balance between the integrated processes of protein folding, degradation and translocation. The Networking of Chaperones by Co-chaperones describes how the function of the major molecular chaperones is regulated by a cohort of diverse non-client proteins, known as co-chaperones. The second edition includes the current status of the field and descriptions of a number of novel co-chaperones that have been recently identified. This new edition has a strong focus on the role of co-chaperones in human disease and as putative drug targets. The book will be a resource for both newcomers and established researchers in the field of cell stress and chaperones, as well as those interested in cross-cutting disciplines such as cellular networks and systems biology.

Co-chaperones are important mediators of the outcome of chaperone assisted protein homeostasis, which is the dynamic integration of the processes of protein folding, degradation and translocation to ensure that cellular function is finely tuned in space and time. This third edition of the book The Networking of Chaperones by Co-chaperones describes how the function of the major molecular chaperones is regulated by co-chaperones, a diverse cohort of non-client proteins. Since the second edition was released, not only has knowledge deepened on how co-chaperones act as nodes to network and functionalise chaperones, but an understanding of their broader biological function has started to emerge. The third edition provides new and updated chapters highlighting recent developments and emerging themes on co-chaperones, such as their extracellular functions, their role in human disease and their status as putative drug targets. The book is a useful resource for both newcomers and established researchers in the field of cell stress and chaperones, as well as those interested in cross-cutting disciplines such as cellular networks and systems biology.

We are extremely happy to present the reader this book containing a summary of a well-known research field, the phenomenon of cellular stress defense from two new angles: networks and membranes. The volume starts with an introduction to the concept of molecular chaperones in their original sense: R. John Ellis, the founder of the chaperone concept describes chaperones as mediators of correct assembly and/or misassembly of other macromolecular complexes. This sets the tone of the book, where later chapters give detailed examples of the richness of chaperone ac tion by hundreds of other proteins and membrane structures. The reader will learn the role of chaperone classes such as Hsp27 or Hsp90, the action of highly organized chaperone networks in various cellular compartments such as the ER or mitochondrial/ER networks as well as the molecular details of the signaling mechanisms leading to chaperone induction during stress. Various special stress defense mechanisms against oxidative stress or dryness will also be covered. Membranes comprise a surprising mixture of stability and dynamics in the cell. Their role in the regulation of the stress response has been accepted only slowly in the field. Two chapters summarize this important aspect of the stress response showing the importance of membrane hyperstructures, lipid species composition, protein/ membrane interactions and cold adaptation.

Proteostasis written by Nidhi Puranik and has been published by Springer Nature this book supported file pdf, txt, epub, kindle and other format this book has been release on 2025-07-12 with Science categories.
This book explores the role of proteostasis in neurodegenerative disorders, focusing on protein homeostasis in long-lived, nondividing cells like neurons. It examines how disruptions in proteostasis, influenced by factors such as oxidative stress, aging, and environmental stress, contribute to diseases like Alzheimer’s, Parkinson’s, and amyotrophic lateral sclerosis. The book begins with an overview of the epidemiology and management of neurodegenerative diseases. Subsequent chapters delve into the mechanisms governing proteostasis, highlighting the ubiquitin-proteasome system, molecular chaperones, and co-chaperones in protein folding and degradation. It further explores how impairments in these pathways contribute to neuroinflammation and disease progression. By providing insights into these cellular processes, the book aims to enhance understanding and support the development of potential therapeutic strategies.
