Understanding the underlying mechanisms of how axons and dendrites develop is a fundamental problem in neuroscience and a main goal of research on nervous system development and regeneration. Previous studies have provided a tremendous amount of information on signaling and cytoskeletal proteins regulating axonal and dendritic growth and guidance. However, relatively little is known about the relative contribution and role of cytoskeletal dynamics, transport of organelles and cytoskeletal components, and force generation to axonal elongation. Advancing the knowledge of these biomechanical processes is critical to better understand the development of the nervous system, the pathological progression of neurodegenerative diseases, acute traumatic injury, and for designing novel approaches to promote neuronal regeneration following disease, stroke, or trauma. Mechanical properties and forces shape the development of the nervous system from the cellular up to the organ level. Recent advances in quantitative live cell imaging, biophysical, and nanotechnological methods such as traction force microscopy, optical tweezers, and atomic force microscopy have enabled researchers to gain better insights into how cytoskeletal dynamics and motor-driven transport, membrane-dynamics, adhesion, and substrate rigidity influence axonal elongation. Given the complexity and mechanical nature of this problem, mathematical modeling contributes significantly to our understanding of neuronal mechanics. Nonetheless, there has been limited direct interaction and discussions between experimentalists and theoreticians in this research area. The purpose of this Frontiers Research Topic is to highlight exciting, and important work that is currently developing in the fields of neuronal cell biology, neuronal mechanics, intracellular transport, and mathematical modeling in the form of primary research articles, reviews, perspectives, and commentaries.

Phosphoinositides play a major role in cellular signaling and membrane organization. During the last three decades we have learned that enzymes turning over phosphoinositides control vital physiological processes and are involved in the initiation and progression of cancer, inflammation, neurodegenerative, cardiovascular, metabolic disease and more. In two volumes, this book elucidates the crucial mechanisms that control the dynamics of phosphoinositide conversion. Starting out from phosphatidylinositol, a chain of lipid kinases collaborates to generate the oncogenic lipid phosphatidylinositol(3,4,5)-trisphosphate. For every phosphate group added, there are specific lipid kinases – and phosphatases to remove it. Additionally, phospholipases can cleave off the inositol head group and generate poly-phosphoinositols, which act as soluble signals in the cytosol. Volume II extends into the role of phosphoinositides in membrane organization and vesicular traffic. Endocytosis and exocytosis are modulated by phosphoinositides, which determine the fate and activity of integral membrane proteins. Phosphatidylinositol(4,5)-bisphosphate is a prominent flag in the plasma membrane, while phosphatidylinositol-3-phosphate decorates early endosomes. The Golgi apparatus is rich in phosphatidylinositol-4-phosphate, stressed cells increase phosphatidylinositol(3,5)-bisphosphate, and the nucleus has a phosphoinositide metabolism of its own. Phosphoinositide-dependent signaling cascades and the spatial organization of distinct phosphoinositide species are required in organelle function, fission and fusion, membrane channel regulation, cytoskeletal rearrangements, adhesion processes, and thus orchestrate complex cellular responses including growth, proliferation, differentiation, cell motility, and cell polarization.

The term polarity in a biological context is used to describe an asymmetry in morphology and distribution of molecules. In neurons, their complex shape with typically one axon and several dendrites reflects this asymmetry. Although neurons assume many different shapes and sizes they always maintain these two domains, which are essential for neuronal function. In the most simple view, neurons use their axon to transmit signals over long distances due to its capacity to extend to enormous lengths. Dendrites, on the other hand, are shorter and receive and integrate signals from different locations. The selection of the site where the axon and dendrites initially emerge during embryonic development is a tightly regulated event, eventually important for the correct formation of neuronal circuits, and disturbances of these processes can have pathological consequences due to circuit malformation. An important question is which mechanisms neurons utilize to specify the sites where axonal and dendrite outgrowth occurs and how their identities are maintained during and after development. The formation of these functionally diverse domains is the result of polarized differences of membrane and protein delivery, mitochondria transport, actin dynamics and microtubule stability. However how and in which temporal order all those events which coordinate the selection and maintenance of axons and dendrites is still under investigation. This selection of articles shall highlight new findings, which help to unravel all molecular and cellular events important for neuronal polarity establishment and maintenance.

Extracellular ATP is currently recognized as one of the most widely distributed neurotransmitters and neuromodulators in the peripheral and central nervous system. ATP-gated P2X receptors are expressed by neurons, glial and many other non-neuronal cells and represent an attractive target for therapeutic interventions. Diverse molecular and cellular mechanisms have been identified for P2X receptor functioning, including the ability to enlarge the size of the ion pore associated with the release of several key immune molecules. A major recent breakthrough was the determination of the X-ray crystal structures of zebrafish P2X4 receptor in ATP-bound and ATP-free states. The P2X receptor research field is rapidly growing, as evidenced by the almost 2000 papers published in the last 5 years. However, despite the fundamental signalling function of extracellular ATP in the nervous system, the widespread roles of P2X receptors have not been widely elucidated and presented in textbooks. In this volume of papers we aim to gather a collection of high quality papers, detailing the latest insights from the most accomplished international P2X receptor researchers. Importantly, basic research into P2X receptors has a strong translational impact and our collection of articles could be a valuable guide for the development of new pharmacological and biotechnological tools addressing the function of P2X receptors.
