Time-resolved fluorescence spectroscopy is widely used as a research tool in bioch- istry and biophysics. These uses of fluorescence have resulted in extensive knowledge of the structure and dynamics of biological macromolecules. This information has been gained by studies of phenomena that affect the excited state, such as the local environment, quenching processes, and energy transfer. Topics in Fluorescence Spectroscopy, Volume 4: Probe Design and Chemical Sensing reflects a new trend, which is the use of time-resolved fluorescence in analytical and clinical chemistry. These emerging applications of time-resolved fluorescence are the result of continued advances in laser detector and computer technology. For instance, pho- multiplier tubes (PMT) were previously bulky devices. Miniature PMTs are now available, and the performance of simpler detectors is continually improving. There is also considerable effort to develop fluorophores that can be excited with the red/ne- infrared (NIR) output of laser diodes. Fluorescence spectroscopy has traditionally found wide application in bio chemistry and cell biology. Since there are relatively few naturally occurring fluorescent biomolecules, fluorescence spectroscopy offers a combination of great specificity and sensitivity. Historically, these features have been ex ploited with great success utilizing both intrinsic and extrinsic probes. Re cent applications have built upon these traditional strengths and have re sulted in the development of new instrumental techniques, novel and convenient fluorescent probes, and a deeper, theoretical understanding of fundamental processes. Frequently, fluorescence techniques are tailored to attack a specific biological problem. These new methods in turn produce new physical situations and phenomena which are often of interest to the physical chemist. Thus, progress in one area stimulates renewed interest in other areas. The goal of this book is to provide detailed monographs on the use of fluorescence to investigate problems at the forefront of biochemistry and cell biology. This book is not meant to be a comprehensive survey but rather to highlight areas of recent developments. It is designed to be readable to the novice and yet provide sufficient detail for the expert to keep abreast of recent developments. The book is organized so that it proceeds from simple biochemical sys tems to more complex cell biological ones. Fluorescence is more and more widely used as a tool of investigation, analysis, control and diagnosis in many fields relevant to physical, chemical, biological and medical sciences. New technologies con tinuously emerge thanks to the progress in the design of light sources (e.g. laser diodes), detectors (3D, 4D) and compact ultrafast elec tronic devices. In particular, much progress has been made in time resolved fluorescence microscopy (FUM: Fluorescence Lifetime Imaging Microscopy; FCS: Fluorescence Correlation Spectroscopy). Furthermore, the sensitivity now allows one to detect a single mole cule in the restricted field of a confocal microscope, which actually offers the possibility to study phenomena at a molecular level. The development of new fluorescent probes is still a necessity. In particular, the growing use of lasers implies high resistance to photo degradation. Fluorescence emission at long wavelengths is also a distinct advantage. Furthermore, in vivo inclusion of new fluorescent aromatic residues in proteins offer new potentialities in biology. of ions and molecules is Fluorescence-based selective detection still the object of special attention. Considerable effort is being made in the design of supramolecular systems in which the recognition event is converted into a fluorescence signal easily detected. New fluorescent sensors for clinical diagnosis and detection of pollutants in atmosphere and water are extensively developed. All these developments justify the regular publication of books giving the state-of-the-art of the methods and applications of fluo rescence spectroscopy.

Fluorescence spectroscopy continues its advance to more sophisticated methods and applications. As one looks over the previous decades, its appears that the first practical instruments for time-resolved measurements appeared in the 1970’s. The instrumentation and analysis methods for time-resolved fluorescence advanced rapidly throughout the 1980’s. Since 1990 we have witnessed a rapid migration of the principles of time-resolved fluorescence to cell biology and clinical appli- tions. Most recently, we have seen the introduction of multi-photon excitation, pump-probe and stimulated emission methods for studies of biological mac- molecules and for cellular imaging. These advanced topics are the subject of the present volume. Two-photon excitation was first predicted by Maria Goppert-Mayer in 1931, but was not experimentally observed until 1961. Observation of two-photon excitation required the introduction of lasers which provided adequate photon density for multi-photon absorption. Since the early observations of two-photon excitation in the 1960s, multi-photon spectroscopy has been limited to somewhat exotic applications of chemical physics, where it is used to study the electronic symmetry of small molecules. Placing one’s self back in 1980, it would be hard to imagine the use of multi-photon excitation in biophysics or cellular imaging.
