Wide and fascinating is the field of research on tryptophan, a most versatile amino acid, transformed, as it is, in our organism into many biologically active substances. This volume contains the proceedings of the Eighth International Meeting on Tryptophan Research, held at the University of Padova, Padova, Italy, from June 25 to 29 1995, under the auspices of the University of Padova, National Research Council, Italian Chemical Society-Division of Pharmaceutical Sciences, Region ofthe Veneto and City of Padova. The meeting was held in Padova to commemorate Prof. Luigi Musajo twenty years after his death and the editors dedicate this book to him in recognition of his pioneering work in tryptophan metabolism. Prof. Osamu Hayaishi delivered the Musajo Memorial Award Lecture: Tryptophan oxygenase. and sleep. Figure I shows the ISTRY President Prof. Simon N. Young presenting the Musajo Memorial Medal to Prof. Hayaishi during the Opening Ceremony. Two hundred scientists from twenty two countries participated in the meeting. These proceedings contain 121 papers encompassing a variety of topics and disciplines.

This volume contains the proceedings of the Ninth Meeting of the "International Study Group for Tryptophan Research" (lSTRY), held at the University of Hamburg, Germany, from October 10 to 14, 1998. At this meeting the recent developments in the field of tryptophan research were presented by leading researchers from all over the world in 81 oral and 48 poster contri butions. Research on tryptophan and its derivatives provides an inexhaustible subject. At the conference we tried to compose a multifacetted picture of the recent investiga tions through contributions from the major disciplines involved. Thus, we tried to strike a balance between basic research topics and clinical, nutritional or industrial applica tions. We offered workshops on tryptophan (in sleep and mood), melatonjn, IDO-acti vation and the eosinophilia-myalgia syndrome (EMS) as a platform for intensive discussion for the participants. In these proceedings many contributions are multidisciplinary and have practical or theoretical implications for different research fields. Hence, we have organized this volume in nine main chapters according to basic disciplines and subjects. We are aware that this classification is artificial, but we hope that it is the best compromise for contributors and readers.

Tryptophan is not only an essential amino acid but also an effective signaling molecule as an indicator of amino acid sufficiency. Serotonin is biosynthesized from the essential amino acid tryptophan. The first chapter of this book deals with the effects of tryptophan as well as some strategies of treatments that modify serotonin levels on cognitive processes during aging and neurodegenerative diseases. The changes of tryptophan metabolism under immunological challenges and physiological conditions like stress are examined as well. In the third chapter, the authors illustrate the key role of L-tryptophan in metabolic functions with special emphasis on the alterations of circadian rhythms. The remaining chapters of the book focus on new research findings on L-tryptophan synthesis from D-serine; the complex experimental and theoretical studies of the tryptophan molecule in its collisions with low and high energy electrons; and finally, a commentary on a novel tryptophan-selective amino acid transporter that is functionally coupled to ID01-dependent signaling pathways.

Tryptophan is a rate-limiting essential amino acid and a unique building block of peptides and proteins. This largest amino acid serves as the precursor for the important endogenous indoleamines serotonin, N-acetylserotonin, and melatonin that act as neurotransmitters, neuromodulators, and neurohormones. Kynurenic acid is the most potent endogenous antiexitotoxic agent. Other highly relevant pathways of tryptophan are the reversible transamination to indole-3-pyruvate with formation related indolic acids that act as potent antioxidant agents. Tryptophan metabolites, such as melatonin, and structurally related agents, such as indole-3-propionic acid, act as potent catalytic antioxidants and bioenergetic agents that facilitate regeneration and protection against stress and aging. Several indole compounds act as uremic toxins since these agents can induce radical formation that is associated with enhanced oxidative stress and damage. The exploration of the effects of these protective and toxic tryptophan derived agents has revealed important molecular mechanisms and mediators of adaptation and aging. Research on tryptophan in nutrition and health can facilitate the development of new approaches to extend human health and life span. Amino acids are the building blocks of life that enable repair, as well as recycling and regeneration. Research on nutrients like amino acids, such as tryptophan and its metabolites, as well as peptides and proteins, or extracts containing this molecular metabolism modifiers can improve health. Research into the indololome is a new emerging and rapidly growing field of utmost relevance to science and society.
