Natural killer cells are an integral component of the innate immune response that can quickly detect and eliminate infected or transformed cells without requiring prior sensitization. It has been demonstrated that NK cells play an important role in tumor immunosurveillance; however, the mechanisms regulating NK cell anti-tumor activity are still not well defined. The work presented in this thesis examines the effects of E1A-mediated transformation, interleukin-2I (IL-21) treatment, and DAP 12-mediated signaling on NK cell-mediated tumor rejection.

Natural killer (NK) cells are immune cells that can eliminate pathogenic microorganisms, aging cells, and tumor cells. State-of-the-art renewal has indicated the feasibility of large-scale preparation of autogenous and allogeneic NK cells from a variety of sources including peripheral blood, umbilical cord blood, placental blood, NK cell lines, and even stem cells such as pluripotent stem cells and hematopoietic stem cells. In the meantime, a range of methodologies has been developed for ex vivo expansion and activation of NK cells such as monolayer culture, coculture with trophoblast cells, bioreactors, cytokine cocktails, and physicochemical irritation. Natural killer (NK) cells are crucial components of the innate immune system and play critical roles in host immunity against viral infections and cancer. NK cells' activity is controlled by the interaction of a wide range of receptors expressed on their surfaces with cell surface ligands. Opposite signals delivered by inhibitory and activating receptors tightly regulate NK cells' cytotoxicity. Natural killer cells can discriminate between normal and cancer cells. NK cells are known to directly recognize and kill malignant cells or induce apoptosis. However, tumor cells have the ability to evade those attacks. The main mechanisms involve the lack of expression or downregulation of the expression of major histocompatibility complex (MHC) class I molecules and secretion of soluble NKG2D ligands by tumor cells. Furthermore, tumors harbor a population of cancer stem cells (CSCs), which can drive tumor progression and therapeutical resistance. This chapter highlights the roles of NK cells in tumor immunosurveillance and their applications for cancer immunotherapy. NK cell biology and function as well as the role of their receptor interactions will be described. We will discuss the therapeutic applications of NK cells in cancer and NK cells targeting CSCs as a promising strategy for cancer therapy.

Natural killer T (NKT) cells are a heterogeneous group of T cells that share properties of both T cells and natural killer (NK) cells. Many of these cells recognize the non-polymorphic CD1d molecule, an antigen-presenting molecule that binds self- and foreign lipids and glycolipids. Upon activation, NK T cells are able to produce large quantities of interferon-gamma, IL-4, and granulocyte-macrophage colony-stimulating factor, as well as multiple other cytokines and chemokines (such as IL-2 and TNF-alpha). NKT cells seem to be essential for several aspects of immunity because their dysfunction or deficiency has been shown to lead to the development of autoimmune diseases (such as diabetes or atherosclerosis) and cancers. NKT cells have recently been implicated in the disease progression of human asthma. The clinical potential of NKT cells lies in the rapid release of cytokines (such as IL-2, IFN-gamma, TNF-alpha, and IL-4) that promote or suppress different immune responses.

The interplay of innate and adaptive antitumor immunity dictates the intensity and outcome of the endogenous anticancer response. Stress-induced molecules on tumor cells trigger innate immune reactions, whereas the processing and presentation of tumor-associated antigens evokes adaptive immune recognition. Innate and adaptive antitumor responses may impact tumor development in different ways. In some cases, endogenous reactions suppress tumor formation, while exerting a selective pressure that fosters the emergence of escape variants. Alternatively, some host responses promote tumor cell growth, invasion, and metastasis through the elaboration of inflammatory mediators and cytokines. Investigations have uncovered unique and overlapping roles for innate and adaptive anti tumor immunity, revealing a complex network of interactions among tumor cells, immune elements, and stromal components within the tumor microenvironment, which together shape the direction, quality, and dynamics of the anticancer response.
