Both qualitative and quantitative resistance mechanisms are important contributors to disease resistance in rice. To identify useful sources of durable resistance for Korean breeding programs, the distribution of rice blast isolates compatible to widely used resistance (R) genes was analyzed. Interactions of 3,747 Korean rice Magnaporthe oryzae isolates with eight monogenic lines, each harboring a major blast R gene, were tested. Lines with R gene Pi-9 and Pi-5 were susceptible to the fewest M. oryzae isolates, and therefore, this gene might be applied for blast resistance in breeding programs in Korea. Six major blast resistance genes were susceptible to more than 60 % of the population, suggesting limited utility of these genes in breeding programs. Quantitative trait loci (QTL)-based resistance is predicted to provide durable and broad spectrum resistance to rice diseases. A candidate gene approach was applied to a population of 164 recombinant inbred lines to identify sources of quantitative resistance. Resistance gene analogs and defense response genes were mapped on the rice chromosomes, and analyzed for their association with blast and bacterial blight resistance QTL. A total of 21 putative QTL for blast resistance were identified on chromosomes 1, 4, 5, 6, 8, 9 and 12. Four putative QTL for bacterial blight resistance were identified on chromosome 3, 5 and 10. Thirteen RGA markers were associated with 11 different QTL on chromosome 1, 5, 8, and 9. The role of one disease resistance QTL associated gene, Os02g39330, encoding a chitinase was investigated for contributing to basal defense responses. RNAi silencing was used to evaluate contributions of the gene for the resistance to Rhizoctonia solani and M. oryzae. Five transgenic lines harboring the silencing construct and which differed in the level of expression of Os02g39330 were screened for responses to R. solani and M. oryzae. The chitinase gene expression levels were inversely correlated with sheath blight disease severity, suggesting a role for this defense gene in resistance to R. solani. Rice blast disease was not affected by silencing Os02g39330. Both qualitative and QTL-based resistances provide valuable sources of disease resistance, and a combination of R gene Pi-9 and QTL harboring the Os02g39330 chitinase may help to stabilize resistance.

Rice blast, caused by the fungal pathogen Magnaporthe grisea, is one of the most destructive rice diseases worldwide and destroys enough rice to feed more than 60 million people annually. Due to high variability of the fungal population in the field, frequent loss of resistance of newly-released rice cultivars is a major restraint in sustainable rice production. In the last few years, significant progress has been made in understanding the defense mechanism of rice and pathogenicity of the fungus. Rice (Oryza sativa L.) is a valuable resource for understanding the complex processes controlling yield and value-added traits. Bacterial blight (BB) is a vascular disease of rice, caused by strains of Xanthomonas oryzae pv. oryzae (Xoo) and provides insight, both practical and basic, into the concepts of susceptibility and resistance. Basic knowledge has been empirically and, more recently, intentionally exploited for broad and durable resistance to the disease. Bacterial blight involves representatives of most classes of resistance genes (R genes) and pathways for basal plant immunity. The study of BB also revealed novelties not observed in other models, possibly due to the long history of rice cultivation and the constant disease pressure. Conspicuous are the recessive R genes that target the notorious type III Transcription Activator-like effectors (TALes) of Xoo. Results indicate that pathogen and host are currently in a battle over a small patch of ground involving TALes function. At the same time, analyses of rice disease physiology are adding to a growing body of knowledge for plant disease processes and to how these processes are intertwined with disease susceptibility. The basic processes of BB present rich targets for the rapid advances in genome editing.

From February 24 -28, 1992 an international symposium on Durability of Disease Resistance was held at the International Agricultural Centre in Wageningen, the Netherlands. The symposium, organized by the Department of Plant Breeding of Wageningen Agricultural University and the Centre for Plant Breeding and Repro duction Research, CPRO-DLO, was part of the DGIS funded programme Durable Resistance in Developing Countries. Without any form of prevention or protection nearly all crops will be seriously or even severely damaged by a range of pathogens. In modern agriculture man has been able to control many if not most pathogens using i) pesticides, ii) phyto sanitary methods such as control of seed and plant material in order to start a crop disease free, iii) agronomic measures such as crop rotation, iv) disease resis tance or combinations of these measures. Over the years the use of pesticides has increased enormously and so did the pro blems associated with pesticide use, such as environmental pollution and building of resistance and tolerance to these pesticides in the pathogens. The use of resis tance too increased strongly over the years and here too problems arose.

The demand for increased food production needs to be met from ever reducing resources of land, water and other environmental constraints. Rice remains the staple food source for a majority of the global populations, but especially in Asia where ninety percent of rice is grown and consumed. Climate change continues to impose abiotic and biotic stresses that curtail rice quality and yields. Researchers have been challenged to provide innovative solutions to maintain, or even increase, rice production. Amongst them, the ‘green super rice’ breeding strategy has been successful for leading the development and release of multiple abiotic and biotic stress tolerant rice varieties. Recent advances in plant molecular biology and biotechnologies have led to the identification of stress responsive genes and signaling pathways, which open up new paradigms to augment rice productivity. Accordingly, transcription factors, protein kinases and enzymes for generating protective metabolites and proteins all contribute to an intricate network of events that guard and maintain cellular integrity. In addition, various quantitative trait loci associated with elevated stress tolerance have been cloned, resulting in the detection of novel genes for biotic and abiotic stress resistance. Mechanistic understanding of the genetic basis of traits, such as N and P use, is allowing rice researchers to engineer nutrient-efficient rice varieties, which would result in higher yields with lower inputs. Likewise, the research in micronutrients biosynthesis opens doors to genetic engineering of metabolic pathways to enhance micronutrients production. With third generation sequencing techniques on the horizon, exciting progress can be expected to vastly improve molecular markers for gene-trait associations forecast with increasing accuracy. This book emphasizes on the areas of rice science that attempt to overcome the foremost limitations in rice production. Plant diseases and pests are a major constraint to agricultural production despite the various measures used to control them. Chemical control, although often e~~ective, may pose environmental hazards and is relatively expensive, especially in developing countries where it may be completely uneconomic. Control through genetically mediated resistance to diseases and pests, is both cheap and environmentally sa~e and at present most diseases and pests o~ staple ~ood crops are controlled through some form of resistance. One of the basic problems in the use of resistance is its ~re quent lack of durability; very often a type of resistance is used that 'breaks down' after a certain period. The temporary nature of this resistance, due to the development of new strains of pest or pathogen able to overcome it, has seriously hindered the improvement o~ the yield potential of many crops as a continuing effort is needed to replace old cultivars who resistance has failed, with new ones. Following Vanderplank's now classical publications (1963, 1968) which differentiated horizontal and vertical resistance, studies on several host-parasite systems have shown that di~ferent types of resistance can be distinguished genetically and epidemiologically, and on the ability o~ the pests or pathogens to adapt to them. A knowledge of how resistance operates at the population level has also opened up possibilities of 'managing' relatively simple resistance types in such a way that a stable host-pathogen system can be pro duced with a minimum of crop loss.

Human population is escalating at an enormous pace and is estimated to reach 9.7 billion by 2050. As a result, there will be an increase in demand for agricultural production by 60–110% between the years 2005 and 2050 at the global level; the number will be even more drastic in the developing world. Pathogens, animals, and weeds are altogether responsible for between 20 to 40 % of global agricultural productivity decrease. As such, managing disease development in plants continues to be a major strategy to ensure adequate food supply for the world. Accordingly, both the public and private sectors are moving to harness the tools and paradigms that promise resistance against pests and diseases. While the next generation of disease resistance research is progressing, maximum disease resistance traits are expected to be polygenic in nature and controlled by selective genes positioned at putative quantitative trait loci (QTLs). It has also been realized that sources of resistance are generally found in wild relatives or cultivars of lesser agronomic significance. However, introgression of disease resistance traits into commercial crop varieties typically involves many generations of backcrossing to transmit a promising genotype. Molecular marker-assisted breeding (MAB) has been found to facilitate the pre-selection of traits even prior to their expression. To date, researchers have utilized disease resistance genes (R-genes) in different crops including cereals, pulses, and oilseeds and other economically important plants, to improve productivity. Interestingly, comparison of different R genes that empower plants to resist an array of pathogens has led to the realization that the proteins encoded by these genes have numerous features in common. The above observation therefore suggests that plants may have co-evolved signal transduction pathways to adopt resistance against a wide range of divergent pathogens. A better understanding of the molecular mechanisms necessary for pathogen identification and a thorough dissection of the cellular responses to biotic stresses will certainly open new vistas for sustainable crop disease management. This book summarizes the recent advances in molecular and genetic techniques that have been successfully applied to impart disease resistance for plants and crops. It integrates the contributions from plant scientists targeting disease resistance mechanisms using molecular, genetic, and genomic approaches.
