Metastasis is the main cause of cancer patient mortality. Local tumor invasion is a key step in metastatic dissemination whereby cancer cells dislodge from primary tumors, migrate through the peritumoral stroma and reach the circulation. This is a highly dynamic process occurring in three dimensions that involves interactions between tumor, stromal cells, and the extracellular matrix. Here we describe the organotypic culture system and its utility to study breast cancer cell invasion induced by cancer-associated fibroblasts. This is a three-dimensional model that reproduces the biochemical and physiological properties of real tissue and allows for investigating the molecular and cellular mechanisms involving tumor and its microenvironment, and their contribution to cancer cell invasion. This system provides a robust, accurate, and reproducible method for measuring cancer cell invasion and represents a valuable tool to improve the mechanistic understanding of the initial steps in metastasis.

Cancer Morbidity and mortality result from invasive and metastatic spread. Currently, no therapies are aimed at the underlying mechanisms that enable this progression due to only nascent recognition of the distinct biology which occurs only during tumor dissemination. Recent advances have highlighted the central role of cell motility during the dynamic and transient process of tumor invasion and metastasis. This book includes state-of-the-art updates by international leaders in these studies. Chapters first present the novel model systems that enable new investigations and insights. Chapters then describe in depth the key processes and molecules that may be therapeutically targeted. Finally, the role of cell motility and its signals is explored in a number of key tumor types. This compilation should be useful to researchers in basic and translational oncology as well as those developing novel agents to prevent tumor invasion and metastasis.

Cell Migration written by Frank Entschladen and has been published by Karger Medical and Scientific Publishers this book supported file pdf, txt, epub, kindle and other format this book has been release on 2010 with Science categories.

Cell migration is a highly complex process which involves several compartments of the cell, including surface receptors, signalling elements and the cytoskeleton. It plays an essential role in embryogenesis, wound healing and inflammatory responses, and a dysregulation of cell movement can cause pathological states such as developmental defects, chronic inflammation, cancer invasion and metastasis. Covering extracellular regulatory signals and intracellular signal transduction pathways as well as the molecular mechanisms of migration in stem cells, leukocytes and tumor cells in the adult human organism, this book summarizes the current state of knowledge about cell migration. In the first part, the major aspects of different migratory cells in health and disease are covered, with special emphasis on T lymphocytes. The second part provides a comprehensive overview of the principal molecular mechanisms of migration such as adhesion receptors, cytoskeletal rearrangements and locomotor force generation, which, together, can be referred to as a cell's 'migrosome'.With contributions by eminent international scientists from different disciplines this book will serve as a valuable resource not only for researchers in cell biology, immunology and oncology, but also for clinicians who wish to learn more about the role of migratory processes in health and disease.

Cell Movement in Health and Disease brings the several scientific domains related to the phenomena together, establishing a consistent foundation for researchers in this exciting field. The content is presented in four main section. The first explores the foundations of Cell Movement, including overviews of cellular structure, signaling, physiology, motion-related proteins, and the interface with the cellular membrane. The second part covers the biological aspects of cellular movement, starting with chemical and mechanical sensing, describing the types of cell movement, mechanics at cell level, cell physiology, collective behavior, and the connections with the extracellular matrix. The following chapters provide an overview of the molecular machinery involved and cell-type specific movement. The third part of the book is dedicated to the translational aspects of cell movement, highlighting the key conditions associated with cell movement dysfunction, like cell invasion in cancer, wound healing, developmental issues, neurological dysfunctions, and immune response. The final part of the book covers key methods and modeling tools for cell movement research, including predictive mathematical models, in vitro and in vivo methods, biophysical and bioinformatics tools. Cell Movement in Health and Disease is the ideal reference for scientists from different backgrounds converging to expand the understanding of this key cellular process. Cellular and molecular biologists will gain a better understanding of the physical principals operating at cellular level while biophysicist and biomedical engineers will benefit from the solid biology foundation provided by the book. Combines Biology, Physics and Modeling of cellular movement in one single source Updated with the current understanding of the field Includes key research methods for cell movement investigation Cover translational aspects of cellular movement
