This book addresses the molecular bases of some of the most important biochemical rhythms known at the cellular level. Clarifying the mechanism of these oscillatory phenomena is of key importance for understanding the origin as well as the physiological function of these rhythms, and the conditions in which simple periodic behaviour transforms into complex oscillations, including bursting and chaos. The approach rests on the analysis of theoretical models closely related to experimental observations. The thalamus is often described as a relay. Typified by sensory pathways, this concept leads to thalamic nuclei being viewed as areas that passively streams information from a single source to the cortex, without affecting the nature of that information. However, diverse intrathalamic connections, the varying synaptic and membrane properties of thalamic neurons and the large number of inputs from non-sensory sources make the idea that the thalamus is just a passive relay unlikely. Furthermore, a large number of thalamic nuclei are not primarily driven by sensory signals nor do they exclusively target the cortex, meaning the thalamus must do more than simply pass sensory signals to the cortex. Finally, there is a wealth of research demonstrating that the thalamus does indeed function in ways that are not captured by the concept of a simple relay. So why, given all of this, is the primary paradigm for describing the thalamus, a relay? There is no doubt that nowadays, biology benefits greatly from mathematics. In particular, cellular biology is, besides population dynamics, a field where tech niques of mathematical modeling are widely used. This is reflected by the large number of journal articles and congress proceedings published every year on the dynamics of complex cellular processes. This applies, among others, to metabolic control analysis, where the number of articles on theoretical fundamentals and experimental applications has increased for about 15 years. Surprisingly, mono graphs and textbooks dealing with the modeling of metabolic systems are still exceptionally rare. We think that now time is ripe to fill this gap. This monograph covers various aspects of the mathematical description of enzymatic systems, such as stoichiometric analysis, enzyme kinetics, dynamical simulation, metabolic control analysis, and evolutionary optimization. We believe that, at present, these are the main approaches by which metabolic systems can be analyzed in mathematical terms. Although stoichiometric analysis and enzyme kinetics are classical fields tracing back to the beginning of our century, there are intriguing recent developments such as detection of elementary biochemical syn thesis routes and rate laws for the situation of metabolic channeling, which we have considered worth being included. Evolutionary optimization of metabolic systems is a rather new field with promising prospects. Its goal is to elucidate the structure and functions of these systems from an evolutionary viewpoint.

This book first reviews the case that brain oscillations not only are important for cognition, as long suspected, but also play a part in the expression of signs and symptoms of neuropsychiatric disorders. The cellular mechanisms of many of the clinically relevant oscillations have been studied by the authors and their colleagues, using in vitro slice methods as well as detailed computer simulations. A surprising insight is that gap junctions between principal neurons play an absolutely critical role in so many types of oscillation in neuronal populations; oscillations are not just the result of properties of individual neurons and their synaptic connections. Furthermore, the way in which gap junctions produce oscillations in the cortex is novel, involving as it does global properties of networks, rather than just the time constants of membrane currents. This insight has implications for therapeutics as well as for our understanding of normal brain functions.
