Up-to-date meeting report of the Else Kr ner-Fresenius Symposium on Adult Stem Cell Aging Adult stem cells are present in most postnatal tissues of mammals. Tissues with high rates of cell turnover depend on the functional capacity of stem cells for lifelong maintenance of tissue homeostasis. Adult stem cells are also required for the regeneration of tissues in response to injury as in, for example, the regeneration of skeletal muscle. In addition to its function in tissue homeostasis and regeneration, adult stem cells can represent the cell type of origin of various types of cancers including leukemia and colorectal cancer. Stem cells are the most long-lived cells in the proliferative compartment of mammalian tissues. Therefore, stem cells have an increased risk of acquiring mutations that could ultimately lead to the transformation of tissue stem cells. This publication presents the current knowledge in the field of stem cell aging, which was discussed at the Else Kr ner-Fresenius Symposium on Advances in Stem Cell Aging in 2011. It will be of special interest to scientists working on stem cell research, aging, regeneration, and cancer as well as physicians and scientists specializing in geriatric medicine, internal medicine, and surgery.

The proper physiological functioning of most eukaryotic cells requires their assembly into multi-cellular tissues that form organized organ systems. Cells of the immune system develop in bone marrow and lymphoid organs, but as the cells mature they leave these organs and circulate as single cells. Antigen receptors (TCRs) of T cells search for membrane MHC proteins that are bound to peptides derived from infectious pathogens or cellular transformations. The detection of such speci?c peptide–MHC antigens initiates T cell activation, adhesion, and immune-effectors functions. Studies of normal and transformed T cell lines and of T cells from transgenic mice led to comprehensive understanding of the mole- lar basis of antigen-receptor recognition and signaling. In spite of these remarkable genetic and biochemical advances, other key physiological mechanisms that par- cipate in sensing and decoding the immune context to induce the appropriate cellular immune responses remain unresolved. TCR recognition is tightly regulated to trigger sensitive but balanced T cell responses that result in the effective elimination of the pathogens while minimizing collateral damage to the host. The sensitivity of TCR recognition has to be properly tempered to prevent unintended activation by self-peptide–MHC complexes that cause autoimmune diseases. It is likely that once the TCR is engaged by a peptide– MHC and TCR signaling begins, additional regulatory mechanisms, involving other receptors, would increase the ?delity of the response.

This book brings together most of the information available concerning two species that diverged 2-3 million years ago. The objective was to try to understand why two sibling species so similar in several characteristics can be so different in others. To this end, it was crucial to confront all data from their ecology and biogeography with their behavior and DNA polymorphism. Drosophila melanogaster and Drosophila simulans are among the two sibling species for which a large set of data is available. In this book, ecologists, physiologists, geneticists, behaviorists share their data on the two sibling species, and several scenarios of evolution are put forward to explain their similarities and divergences. This is the first collection of essays of its kind. It is not the final point of the analyses of these two species since several areas remain obscure. However, the recent publication of the complete genome of D. melanogaster opens new fields for research. This will probably help us explain why D. melanogaster and D. simulans are sibling species but false friends.
