The relationship between metabolic dysfunctions and cancer development has been continuously evolving, informed by research efforts in the fields of epidemiology, endocrinology, cancer biology, chemistry, genomics, metabolomics among others. Metabolism and cancer can intersect at the physiological level, where metabolic disorders and cancer malignancies occur as comorbid or mutually-promoting conditions; or at the molecular level, where metabolic alterations regulate tumorigenesis and oncogenic/tumor-suppressive pathways govern the onset of metabolic syndromes. The increasingly complex spider web connecting metabolic dysfunctions and cancer development reveals both distinctive and shared mechanisms underlying different disease indications. It also points to unique opportunities, either through repurposing existing tools or discovering new strategies, to develop targeted therapies against a wide variety of metabolic and cancerous diseases. In our previous Research Topic: Double-edged Swords: Genetic Factors That Influence The Pathogenesis of Both Metabolic Disease and Cancer , we highlighted ten genetic factors/mechanisms that influence the pathogenesis of both metabolic disease and cancer. The attention it generated to date ( >110,000 article views, >22,000 article downloads, >500 citations) signals a fast-growing interest from scientific communities of different disciplines. We aim to highlight more recent advances to inform a diverse audience working on basic and translational research.

Pediatric oncology and hematopoietic cell transplant (HCT) patients with a critical illness are a particularly challenging population with a unique presentation, diagnosis, and management. One in every three to four children with cancer will require admission to the Pediatric Intensive Care Unit (PICU) during the course of their treatment. Providing optimum care for these patients involves dedicated multidisciplinary collaboration to provide timely and appropriate management. Onco-critical care is a growing field of expertise for the care of this high acuity patient population. Recent work in this field has focused on sepsis, respiratory complications, and multi-organ failure in HCT patients, demonstrating significantly higher morbidity and mortality compared to other critically ill pediatric patients. For the past decade, the HCT/oncology subgroup of the Pediatric Acute Lung Injury and Sepsis Investigators (PALISI) Network has fostered both research and clinical collaborations between oncologists, transplanters, and critical care physicians with the goal to improve outcomes in critically ill oncology and HCT patients. Contributors to this Research Topic will be actively recruited from this network.

One of the major breakthroughs of the last decade in the understanding of energy homeostasis is the identification of a reciprocal control between circadian rhythmicity and cellular metabolism. Circadian rhythmicity is a fundamental endogenous process of almost every organism living on Earth. For instance, the alternation of hunger and satiety is not continuous over 24 h, but is instead structured in time along the light/dark cycle. In mammals, the temporal organization of metabolism, physiology and behavior around 24 h is controlled by a network of multiple cellular clocks, synchronized via neuronal and hormonal signals by a master clock located in the suprachiasmatic nuclei of the hypothalamus. This central circadian conductor in the brain is mainly reset by ambient light perceived by the retina, while secondary circadian clocks in other brain areas and peripheral organs can be reset by meal timing. Chronic disruption of circadian rhythms, as seen in human shift-workers (up to 20% of the active population), has been associated with the development of a number of adverse mental and metabolic conditions. Understanding of the functional links between circadian desynchronization and overall health in animal models and humans, however, is still scarce. Interactions between circadian clocks and metabolism can occur at different levels: the molecular clockwork, internal synchronization via neuro-hormonal signals, or external synchronization via photic or feeding cues. This Research Topic comprises a number of reviews as well as research and methods articles that feature recent advancements in the mechanisms linking circadian clocks with energy metabolism, and the pathophysiological implications of these interactions for metabolic health.
