This book is about melanoma—its biology, immunology, and pathology, as well as the initial use of powerful genomic tools to study its fundamental mole- lar and genetic characteristics. The study of cancer will be profoundly impacted by the Human Genome Project. I would like to discuss some of these changes. The first draft of the human genome sequence was announced in June 2000, and we have just scratched the surface of the changes it will engender in medicine. A relevant question is what are the long-term effects of the Human Genome Project for medicine? I would argue that there are three, and each of these three point toward the view that systems biology will dominate biology and medicine of the 21st century. First, the Human Genome Project introduced a new type of s- ence—discovery science. Discovery science takes a biological system (e. g. , the genome) and defines all of its elements (e. g. , the sequences of the 24 human ch- mosomes). Thus, it creates a rich infrastructure from which the classical hypo- esis-driven science can be done more effectively. The effective integration of discovery- and hypothesis-driven science is a key for systems approaches to bi- ogy and medicine. Second, the Human Genome Project has provided a “periodic table of life.

It has become clear that tumors arise from excessive cell proliferation and a c- responding reduction in cell death. Tumors result from the successive accumulation of mutations in key regulatory target genes over time. During the 1980s, a number of oncogenes were characterized, whereas from the 1990s to the present, the emphasis shifted to tumor suppressor genes (TSGs). It has become clear that oncogenes and tumor suppressor genes function in the same pathways, providing positive and ne- tive growth regulatory activities. The signaling pathways controlled by these genes involve virtually every process in cell biology, including nuclear events, cell cycle, cell death, cytoskeletal, cell membrane, angiogenesis, and cell adhesion effects. Tumor suppressor genes are mutated in hereditary cancer syndromes, as well as somatically in nonhereditary cancers. In their normal state, TSGs control cancer development and p- gression, as well as contribute to the sensitivity of cancers to a variety of therapeutics. Understanding the classes of TSGs, the biochemical pathways they function in, and how they are regulated provides an essential lesson in cancer biology. We cannot hope to advance our current knowledge and to develop new and more effective therapies without understanding the relevant pathways and how they influence the present approaches to therapy. Moreover, it is important to be able to access the powerful tools now available to discover these genes, as well as their links to cell biology and growth control.

I would like to mention that my primary reason for agreeing to put this book together is that, when I was a graduate student, I purchased a copy of Selected Methods in Cellular Immunology by Mishell and Shigii which proved a tremendous help in learning the basics of one-and two- dimensional gel te- niques (and other methods). The cover has long since fallen off, but it still remains one of my most valued reference books for the laboratory. It is my hope that T Cell Protocols: Development and Activation will prove similarly useful to current and future scientists wishing to learn new methods for expl- ing the development and activation of T cells.

The aim of MHC Protocols is to document protocols that can be used for the analysis of genetic variation within the human major histocompatibility complex (MHC; HLA region). The human MHC encompasses approximately 4 million base pairs on the short arm of chromosome 6 at cytogenetic location 6p21. 3. The region is divided into three subregions. The telomeric class I region contains the genes that encode the HLA class I molecules HLA-A, -B, and -C. The centromeric class II region contains the genes encoding the HLA class II molecules HLA-DR, -DQ, and -DP. In between is the class III region, originally identified because it contains genes encoding components of the complement pathway. The entire human MHC has recently been sequenced (1) and each subregion is now known to contain many other genes, a number of which have immunological functions. The study of polymorphism within the MHC is well established, because the region contains the highly polymorphic HLA genes. HLA polymorphism has been used extensively in solid organ and bone marrow transplantation to match donors and recipients. As a result, large numbers of HLA alleles have been identified, a process that has been further driven by recent interest in HLA gene diversity in ethnic populations. The extreme genetic variation in HLA genes is believed to have been driven by the evolutionary response to infectious agents, but relatively few studies have analyzed associations between HLA genetic variation and infectious disease, which has been difficult to demonstrate.

Dendritic Cell Protocols provides chapter and verse for many useful practical approaches to the art of studying dendritic cells. The book gives information on the usual techniques for derivation of human dendritic cells from precursor stem cells, such as monocytes. The last section of the book is devoted to functional aspects of dendritic cells ranging from inf- mation relevant to cell migration to antigen uptake and T cell stimulation. But what is a dendritic cell? The explosion of studies on this cell type over the last few years is breathtaking. We now have so-called “myeloid” and “plas- cytoid” dendritic cells; are they of separate lineages or are they related? There are claims that dendritic cells can be derived from B cells and from granulocytes, as well as from “monocytes. ” Lymphocytes can be switched on or off by dendritic cells. Dendritic cells can preferentially cause Th1 or Th2 responses or some may stimulate B cells directly. When there is so much scope for exploring these new developments, it is particularly important that the data that are established and the practical basis for this work are clearly available, a task that this book fulfils.
