Studies of chemical recognition in biology were initiated about half a century ago with the flrst kinetic data obtained on enzyme catalysis and inhibition. They led to a rather static representation of the recognition process illustrated by the lock and key model that still continues to influence our overall image of recognition and its specificity. In several cases, crystallographic studies of enzyme-substrate complexes have supported this model. Indeed, in a crystallized ligand-enzyme complex, a close fltting is observed between the active center of the enzyme and the functional groups of the ligand. How ever, this does not necessarily result from a direct recognition process between rigid structures, but may result from a progressive adaptation during which the initial struc tures of the enzyme and the ligand are modified (induced-flt mechanism). Recently, a great deal of work has been devoted to the study of recognition in more complex systems such as the replication or the translation machin~ries; clearly, the extraordinary precision of such systems cannot be explained solely in terms of physical matching between enzymes and their substrates. This has led to a noticeable change of perspective in these areas. As a result of the new kinetic viewpoint, one rather focuses on the time-course of the processes, on the kinetic balance between steps of the reaction, on the energy-accuracy relationships and on the strategies which permit the achievement of high precision using relatively error-prone components in an appropriate dynamic interplay.

In this book, the molecular recognition of DNA using small molecules is discussed, with a study of the photochemistry of BrU-labeled DNA. The purposes of the study were to develop small molecules for regenerative medicine, to develop a method to detect the recognition site of small molecules, and to detect the most important biological phenomena using the photochemistry of BrU-labeled DNA. The study began with the design and development of small molecules that can induce pluripotency genes. To deal with the important issue of cell permeability of the original compound, a new analogue of the original with improved gene expression was designed and synthesized. Using the photochemistry of BrU-labeled DNA, crucial biological phenomena such as cooperativity between transcription factors were detected. For the first time, the cooperativity was examined by excess electron transfer assay. DNA was also studied very carefully in order to understand the mechanism of the double-strand break in the UVA micro-irradiation technique. The mechanism of the double strand remained untouched. Nevertheless, the double-strand break mechanism was clearly demonstrated by Hoechst dye, as shown in this book.

Man's mind stretched to a new idea never goes back to its original dimensions Oliver Wendell Holmes Our current understanding of sex and biological differentiation results from the application of three principal experimental approaches to these subjects: those of the physiologist, the biochemist, and the geneticist. These three approaches are illustrated by the materials presented in the chapters of this volume. Chapters 1-5 emphasize conceptualization of developmental processes, describing systems principally from the standpoint of the physiologist. Structures and functions are defined with only occasional reference to specific molecular details. Chapters 6- 10 present the views of the biochemist, attempting to describe functions influ encing or regulating cellular behavior at the molecular level. And Chapters 11- 14 illustrate the approaches of the modem-day geneticist in his attempts to gain a detailed understanding of processes controlling gene expression. While it is possible to delineate these three major sections, each emphasizing a distinct experimental approach, it must be realized that the yield of knowledge increases exponentially with the number of experimental approaches available to the investigator.
