Intracellular junctions provide routes for direct cell-to-cell signalling in both plants and animals. The present volume treats the parallels and differences between such junctions in animals and plants and discusses the most recent methods of examining the physiological functions and regulation of intracellular communication. Strong evidence of both molecular as well as functional simi§ larities between plasmodesmata and gap junctions is increasing. Even more interesting is the discovery that animal gap junction proteins cross-react immunologically with some proteins in plant cells. Thus the molecular construction and function of these crucially important ultrastructural cell components is now open to a concerted research effort to understand how cells, both plant and animal, facilitate and regulate intercellular transport.

The Advanced Study Institute on "Structure, Biogenesis and Dynamics of Biological Membranes, held in Cargese from June 14-26, 1993, has been dealing with four major topics in membrane biochemistry today: lipid dynamics and lipid-protein interactions, protein translocation and insertion, intracellular traffic aud protein structure and folding. The lecturers discussed these topics starting from several disciplines, including biochemistry, cell biology, genetics, and biophysics. This wayan interdisciplinary and very inte~sting view on biological membrane systems was obtained. At first an extensive overview of -mainly biophysical -techniques which can be used to study dynamic processes in membranes was presented. Sophisticated approaches such as ESR and NMR have been applied succesfully to unravel details of specific lipid-protein interactions. x ray analysis provides detailed structural information of several proteins and the possible implications for protein functions. Information obtained this way is complemented by studies on mechanisms and kinetics of protein folding. The latter information is indispensable when discussing protein translocation and insertion: proces:;es in which folding and unfolding play essential roles. Extensive insight was offered in the complicated machinery of phospholipid biosynthesis. In particular, the application of sophisticated genetic techniques has allowed a better understanding of the mechanisms regulating the synthetic machinery and detailed studies on a variety of mutants, lacking one or more of the essential enzymes, have resulted in the beginning of a bL!:

Provided here is a comprehensive examination of the basic and clinical condition of three innovative and promising approaches to cancer therapy, which may support or even substitute chemotherapy: differentiation, immunomodulation, and inhibition of angiogenesis. The study of membrane traffic in reconstituted cell-free systems has generated an unprecedented amount of new information on the biochemistry, molecular biology and genetics of membrane-based molecular events that underly normal and abnormal cellular function. Many of the individual steps have now been isolated and dissected in simple systems that permit detailed molecular analyses of transport mechanisms and their regulation. Reconstituted events of intercompartment transport include inter-membrane recognition, and controlled membrane fusion-fission reactions. Among the many advances is the growi ng awareness of a remarkabl e evolutionary conservation of many of the components involved in the many steps of membrane traffic, this realization has accelerated greatly the pace of progress in the field. This book provides a collection of participant contributions from the 1992 Summer Research Conference, "Mol ecul ar Mechani sms of Membrane Traffi c, " jointly sponsored with NATO by the American Society of Cell Biology. The conference was held May 9-13, at the Airlie Conference Center in the Virginia countryside, near Warrenton. The conference was attended by 158 scientists. A unique feature was the high proportion of young scientists among the participants. Approximately 65% were students, postdoctoral fe 11 ows and young investigators. This volume contains the lectures presented at the NATO Advanced Study Institute (ASI) on "Trafficking of Intracellular Membranes: From Molecular Sorting to Membrane Fusion", held in Espinho, Portugal, from June 19 to June 30,1994. The objective of this Institute was to survey recent developments and to discuss future directions in the rapidly advancing field of membrane cell biology, with particular emphasis on the dynamical properties and intracellular flow of membranes. A wide range of interrelated topics around the central theme of intracellular trafficking of membranes was covered, including lipid flow, membrane fusion, dynamics of membrane components, protein folding and assembly, vesicular transport in membrane biogenesis, exocytosis and endocytosis. A large variety of experimental techniques and systems, including the application of viruses and model systems, to study these processes was also considered. Membrane cell biology is a broad discipline which encompasses many scientific areas including cell biology, biochemistry, biophysics, virology, immunonology and genetics. Indeed, recent advances in the cell biology of membranes could not have been made without this multidisciplinary approach. Significant progress achieved during the last few years in understanding how newly synthesized lipids and proteins find their way to the cell organelles, how molecular sorting and the continuous flow of membranes allow each cellular membrane to maintain its own distinct molecular composition, and, thereby, the individuality of the various intracellular compartments, was discussed in considerable detail in this Institute.

Protein insertion and translocation, intracellular traffic and sorting of membranes and their components, and lipid-protein interactions were the main topics of the Advanced Study Institute on "Molecular Dynamics of Membranes", which was held in June 1995 in Cargese, Corsica, France. The course, co-sponsored by NATO and FEBS, was the fifth in a series that started in 1987 and takes place every two years in the Institut d'Etudes Scientifiques in Cargese. This Institute, ideally situated and fully equiped for this type of scientific meeting has greatly contributed to the great success of the courses. Of course, also the outstanding contributions of a large number of well known scientists and the enthousiastic participation of excellent graduate students and postdocs has given the "Cargese Lectures on Biomembranes" a firm reputation in the scientific community. The present proceedings is more than just a reflection on the information presented in the Course. First of all it contains a number of extensive reviews of specific areas of interest. Noteworthy are the articles dealing with: • the general mechanisms of protein transport, the roles of invariant chain in antigen presentation, protein import and export in E. coli, protein folding and the role of chaperones, chloroplast and mitochondrial protein import, • membrane traffic in general and during mitosis, and with respect to membrane lipids: lipid domain formation, lipases: an extensive review about structure and properties, phospholipase A2 and bioactive lipids, phospholipid transfer proteins, • phospholipid localization and mobility and, finally, new strategies for protein reconstitution.
