This book describes transcription factors in the context of hematopoietic development and reveal mechanistic insights how they orchestrate specific gene expression patterns. During vertebrate hematopoiesis many specialized cell types are formed with vastly different functions such as B cells, T cells, granulocytes, macrophages, erythrocytes and megakaryocytes. To tightly control the enormous proliferative potential of developing blood cells, an intricately balanced signaling and transcription network has evolved that ensures that the different cell types are formed at the right time and in the right numbers. Intricate regulatory mechanisms ensure that blood cells function properly and have a determined life span. Moreover, in the adaptive immune system, long-lived memory cells have evolved that ensure that when pathogens have been seen once they will never cause a problem again. In this book we will therefore make a journey from asking how more primitive organisms use the epigenetic regulatory machinery to balance growth with differentiation control towards digging deep into what controls the function of specialized cells of the human immune system. We will first discover that flies make blood but exist without blood vessels, why fish make blood cells in the kidney and which precise genetic circuitries are required for these developmental pathways. We will then learn the regulatory principles that drive the differentiation of mature blood cells from stem cells and what controls their function in mammals. In the process, we will find out what unites hematopoietic stem cells and endothelial cells. Finally, we will shed light on the molecular mechanisms that either alter hematopoietic cell differentiation or lead to the development of cells with impaired function.

The title "Stem Cells from Cord Blood, In Utero Stem Cell Develop ment, and Transplantation-Inclusive Gene Therapy" suggests that more than one topic is combined in one workshop. Indeed, at first glance the recovery of stem cells from cord blood has to be seen as separate from the attempts to achieve effective in utero therapy by stem cell trans plantation, because the first issue deals with an innovative stem cell source as an alternative to bone marrow, which is already spreading rapidly in medical practice, whereas the second topic is still strictly ex perimental and only investigated in medical centers with the appropri ate background. It is, however, not only justified, but helpful to com bine the two topics in one workshop and consequently to cover them in the same volume of the Ernst Schering Research Foundation Work shop series, because they are intimately related and both based on the new insights into the biology of stem cells. Explanatory Particularism in Scientific Practice offers a novel community-centric account of scientific explanation. On this view, explanations are products of collaborative activity in particular communities. Philosophers of science studying explanation have traditionally seen their task as analyzing the common or fundamental core of explanations across the sciences. Melinda Bonnie Fagan takes the opposite view: diversity of explanations across the sciences is a basic feature of scientific practice. A scientific community produces explanations that advance understanding of some target of interest, but just what features advance understanding, and what understanding amounts to in practice, varies widely over time and across scientific communities. This particularist approach brings new problems and questions to the fore, especially concerning interdisciplinarity: how (if at all) do explanation and understanding get beyond the boundary of a particular community? The particularist account also has implications bearing on the nature of understanding, the unity of science, objectivity, and science-society relations. The argument is elaborated using detailed case studies of explanatory model connection, or lack thereof: immunology and epidemiology models in the COVID-19 pandemic and the explanatory ambitions of systems biology, using the example of stem cell development.
