This volume explores the quickly evolving field of Purinergic signaling, and examines how receptors for ATP and other nucleotides, and receptors for adenosine, act in neuronal transmission, control of synaptic activity, proliferation, differentiation and cell death regulation in the CNS. This book focuses on the participation of purinergic receptors and ectonucleotidases, degrading ATP into adenosine, in embryonic and adult neurogenesis in vitro and in vivo as well as in synaptic transmission and pathophysiology. Further, the chapters discuss varying brain diseases, including Parkinson’s, and Alzheimer’s disease, autism, mood disorders and epilepsy, as well as brain tumors, in the context of purinergic signaling and its clinical aspects. The development of purinergic receptor agonists is also an important issue of this book. This book provides a critical review of the current state of science and will be useful for both scientists and students who are or would like to get involved in this area. Furthermore, this book addresses neuroscientists, physician and professionals from the industry, who would like to update themselves in this exciting and rapidly growing field of neuroscience.

In the central nervous system, extracellular matrix (ECM) molecules, including hyaluronic acid, chondroitin and heparan sulfate proteoglycans, tenascins, reelin and agrin, along with their remodelling enzymes, such as neurotrypsin, neuropsin, plasminogen activators, and metalloproteinases, are secreted by neural and non-neural cells into the extracellular space to form the ECM and signal via ECM receptors. Despite recent advances in the ECM field, the importance of neural ECM for physiological and pathological processes is currently less widely recognized than that of other CNS elements. This book will enlighten recent progress in our understanding of mechanisms by which neural ECM, its receptors and activity-dependent ECM remodeling regulate neural development, synaptic plasticity, and contribute to pathological changes in the brain. In the first part, the roles of ECM signaling and proteolytic modification of ECM in neurogenesis, neural migration, axonal pathfinding, synaptogenesis, synaptic and homeostatic plasticity will be discussed. The second part will focus on the emerging ECM-dependent mechanisms associated with CNS injury, epilepsy, neurodegenerative and neuropsychiatric diseases. For further development of neural ECM field, a very important contribution is the third part of the book, which is devoted to neural ECM-targeting tools and therapeutics. The concluding fourth part will highlight advances in development of artificial ECM and ECM-based systems suitable for multisite recording and stimulation of neural cells. Authors are the leading experts in the field of brain extracellular matrix in health and disease Book covers the most important aspects of brain extracellular matrix in health and disease Interesting for both scientists and clinicians

Cyclin Dependent Kinase 5 provides a comprehensive and up-to-date collection of reviews on the discovery, signaling mechanisms and functions of Cdk5, as well as the potential implication of Cdk5 in the treatment of neurodegenerative diseases. Since the identification of this unique member of the Cdk family, Cdk5 has emerged as one of the most important signal transduction mediators in the development, maintenance and fine-tuning of neuronal functions and networking. Further studies have revealed that Cdk5 is also associated with the regulation of neuronal survival during both developmental stages and in neurodegenerative diseases. These observations indicate that precise control of Cdk5 is essential for the regulation of neuronal survival. The pivotal role Cdk5 appears to play in both the regulation of neuronal survival and synaptic functions thus raises the interesting possibility that Cdk5 inhibitors may serve as therapeutic treatment for a number of neurodegenerative diseases.

Immunomodulatory Biomaterials written by Stephen F. Badylak and has been published by Woodhead Publishing this book supported file pdf, txt, epub, kindle and other format this book has been release on 2021-07-30 with Medical categories.

Biomaterials have existed for millennia as mechanical replacement structures following disease or injury. Biomaterial design has changed markedly from structural support with an “inert immune profile as the primary objective to designs that elicit an integrative local tissue response and a pro-repair immune cell phenotype. Immunomodulatory Biomaterials: Regulating the Immune Response with Biomaterials to Affect Clinical Outcome offers a single, comprehensive reference on biomaterials for modulation of the host response, for materials scientists, tissue engineers and those working in regenerative medicine. This book details methods, materials and strategies designed to regulate the host immune response following surgical implantation and thus facilitate specific local cell infiltration and tissue deposition. There has been a dramatic transformation in our understanding of the role of the immune system, both innate and adaptive; these changes include recognition of the plasticity of immune cells, especially macrophages, cross-talk between the immune system and stem cells, and the necessity for in situ transition between inflammatory and regulatory immune cell phenotypes. The exploitation of these findings and the design and manufacture of new biomaterials is occurring at an astounding pace. There is currently no book directed at the interdisciplinary principles guiding the design, manufacture, testing, and clinical translation of biomaterials that proactively regulate the host tissue immune response. The challenge for academia, industry, and regulatory agencies to encourage innovation while assuring safety and maximizing efficacy has never been greater. Given the highly interdisciplinary requirements for the design, manufacture and use of immunomodulatory biomaterials, this book will prove a useful single resource across disciplines. Holistically covers the design, manufacture, testing, and clinical translation of biomaterials that proactively regulate the host tissue immune response Provides a single reference for understanding and utilizing the host response in biomaterials design An international collaboration of leading researchers in the field offering a novel insight into this fast-growing area

Hydrocephalus is one of the most common diseases of pediatric and adult neurosurgery. With the introduction of modern neurosurgical procedures, this disease has become a life-long problem. Even with optimal treatment, there is still significant morbidity and mortality along with a significant cost to the medical system. This has caused patients and their families to demand improvements in treatments and forced clinicians to evaluate their treatments in large consortiums while utilizing both genetics and technology to improve outcomes or avoid placement of shunt all together. This text is designed to present the current treatments for hydrocephalus across the lifespan. The foundation for understanding cerebral spinal fluid (CSF) abnormalities begins with the understanding of physiology and pathogenesis of disease. These chapters are written by published experts in the field and detail the significant advances in the detection of CSF abnormalities. This section will discuss the current advances in imaging and current research in biomarkers for both pediatric and adult patients. We will then systematically discuss the treatment of both pediatric and adult CSF disorders. These will be broken down by cause, since the physiology of each can be different. We will end the book with a discussion both of the technological advances and a discussion of consortiums and how they have advanced treatment of this chronic disease.

Traumatic brain injury (TBI) remains a significant source of death and permanent disability, contributing to nearly one-third of all injury related deaths in the United States and exacting a profound personal and economic toll. Despite the increased resources that have recently been brought to bear to improve our understanding of TBI, the development of new diagnostic and therapeutic approaches has been disappointingly slow. Translational Research in Traumatic Brain Injury attempts to integrate expertise from across specialties to address knowledge gaps in the field of TBI. Its chapters cover a wide scope of TBI research in five broad areas: Epidemiology Pathophysiology Diagnosis Current treatment strategies and sequelae Future therapies Specific topics discussed include the societal impact of TBI in both the civilian and military populations, neurobiology and molecular mechanisms of axonal and neuronal injury, biomarkers of traumatic brain injury and their relationship to pathology, neuroplasticity after TBI, neuroprotective and neurorestorative therapy, advanced neuroimaging of mild TBI, neurocognitive and psychiatric symptoms following mild TBI, sports-related TBI, epilepsy and PTSD following TBI, and more. The book integrates the perspectives of experts across disciplines to assist in the translation of new ideas to clinical practice and ultimately to improve the care of the brain injured patient.
