Based on the widespread but well-ordered brain tissue loss that occurs with healthy aging into senescence, this work presents a novel magnetic resonance imaging (MRI)-based biomarker, which identifies normal and abnormal aging-related brain atrophy. The novel BrainAGE approach is based on a database of structural MRI data, aggregating the complex, multidimensional aging patterns across the whole brain to one single value, i.e. the estimated brain age. Consequently, subtle deviations in "normal" brain atrophy can be directly quantified in terms of years by analyzing one standard MRI scan per subject. Various neuro-degenerative diseases - especially Alzheimer's disease (AD) - are widely linked to advanced brain aging. The BrainAGE approach is applied to identify advanced brain aging in subjects with mild cognitive impairment and AD, to predict conversion to AD, to relate individual BrainAGE scores with disease severity and prospective worsening of cognitive functions. Furthermore, BrainAGE identifies various risk factors of pathological brain aging that may precede the onset of clinical symptoms (e.g., diabetes mellitus type 2, metabolic syndrome).

Episodic memory (EM) is well known to decline with age, and is especially affected in Alzheimer's disease (AD). Notably, however, EM declines are heterogeneous even among individuals with AD, and research indicates that there are a number of important risk and protective factors influencing the patterns and trajectories of these declines. One factor shown to influence aging EM is metamemory, which plays an essential role in the strategies used for encoding and retrieval of information, and the control of memory output. In general, better metamemory is associated with better memory performance, with most studies showing age-related metamemory declines that are worse in AD. However, studies also show that metamemory can be improved, even in those with more advanced AD. The main aim of the current study was to examine the relationships between EM, metamemory, and aging by comparing performance of healthy older and younger adults during an EM learning and recognition task. Measures of metamemory included the Memory Functioning Questionnaire (MFQ), as well as analysis of retrospective confidence judgments and mnemonic strategy use during the EM task. A known risk factor for AD as well as age-related EM declines is possession of an e4- allele of the apolipoprotein-E (ApoE) gene. Importantly, research has shown that carriers of the e4-allele display EM declines beyond those of normal aging, even when AD does not manifest. A second aim of the current study was to examine the effects of apoE genotype on the relationship between aging EM and metamemory. Studies have shown olfactory measures to be useful as early indicators of declines related to AD and apoE. Thus, the current study used an EM odor-learning and memory task, the California Odor Learning Test (COLT), to compare the effects of age, apoE genotype, and metamemory on EM task performance. Ability to detect odors was assessed and controlled for. Participants included 16 older adults (8 ApoE-e4 positive, 8 ApoE-e4 negative), and 25 younger adults (6 ApoE-e4 positive, and 13 ApoE-e4 negative). Age and ApoE groups did not significantly differ on any demographic variables. Results indicated that younger adults performed significantly better than older adults on all measures of recall consistency and recognition memory, as well as on all measures of metamemory abilities with the exception of self-reported use of mnemonics. Significant interaction effects between age and apoE genotype on short and long delay cued recall were found. As predicted, significant positive correlations were found among the recognition and recall measures of the COLT, among the task-based metamemory measures, as well as among the MFQ scales. These findings support the positive relationship between memory performance and metamemory abilities among both healthy older and younger adults, and further this idea to include the positive relationship of online mnemonic strategy use to both memory performance and metamemory abilities. For older adults, better metamemory has been associated with higher quality of life, while poor metamemory can produce serious dayto- day consequences for the individual and those they interact with, negatively affecting judgment and decision-making abilities. Thus, research on aging metamemory remains particularly important, and may help explain the heterogeneity seen in aging EM.

ABSTRACT: In a majority of studies, the apolipoprotein E-[epsilon]4 allele (apoE-[epsilon]4) has been shown to be a significant risk factor for Alzheimer's disease (AD). However, the nature of the association between apoE-[epsilon]4 and cognition in non-demented persons remains unresolved. Some studies have demonstrated that apoE-[epsilon]4 positive individuals perform significantly worse on neuropsychological tests compared to non-apoE-[epsilon]4 individuals, while other studies have reported that apoE-[epsilon]4 is not associated with cognitive impairment in normal aging. Neurobiological studies using animal and human tissue samples suggest that apoE isoforms and protein levels may affect neuronal growth/plasticity and the accumulation of neuropathological changes of the type seen in AD. However, the association between apoE plasma levels and cognition within apoE genotype groups has largely been unexplored. In this study, 1474 non-demented participants from the Framingham Study offspring cohort (mean age = 62.1, SD = 9.5, range = 36-88) were administered a neuropsychological test battery. Participants were divided by apoE genotype: 207 apoE-[epsilon]2 ([epsilon]2/[epsilon]2, [epsilon]2/[epsilon]3), 962 apoE-[epsilon]3 ([epsilon]3/[epsilon]3), and 305 apoE-[epsilon]4 ([epsilon]4/[epsilon]4, [epsilon]3/[epsilon]4), sex, and age ([less than]70 and [greater than or equal to]70). Participants were also divided by the presence or absence of the apoE-[epsilon]4 allele. For a subset of these individuals (n = 1145), apoE plasma levels were also obtained. Results revealed that the apoE-[epsilon]4 group performed significantly worse than the apoE-[epsilon]3 and apoE-[epsilon]2 groups on tests of verbal short-term memory and attention, despite the fact that pre-morbid intellectual function was superior in the apoE-[epsilon]4 group. When examining apoE plasma levels, results showed that high apoE plasma level in apoE-[epsilon]4 participants was associated with poorer cognitive performance. A statistically non-significant trend of better cognitive performance with elevated apoE plasma level was evidenced in apoE-[epsilon]3 individuals. These findings suggest that apoE-[epsilon]4 affects cognition in non-demented persons. This effect was more robust in females than males. Results also demonstrate that apoE plasma levels may influence cognition differently in apoE-[epsilon]3 and apoE-[epsilon]4 individuals.

This book collects and reviews, for the first time, a wide range of advances in the area of human aging biomarkers. This accumulated data allows researchers to assess the rate of aging processes in various organs and systems, and to individually monitor the effectiveness of therapies intended to slow aging. In an introductory chapter, the editor defines biomarkers of aging as molecular, cellular and physiological parameters that demonstrate reproducible changes - quantitative or qualitative - with age.
