Multiple Myeloma written by Paul G. Richardson and has been published by Remedica this book supported file pdf, txt, epub, kindle and other format this book has been release on 2004 with Medical categories.

Multiple myeloma is the second most common hematologic malignancy, affecting 15,000 patients per year in the United States. Despite the advent of high-dose chemotherapy with stem cell transplantation, multiple myeloma remains incurable, with approximately 12,000 deaths per annum recorded in the US from the disease. Over the last 10 years, there has been a dramatic increase in our understanding of the biology of multiple myeloma, which has provided insights into mechanisms of cytotoxic resistance, both as inherent characteristics of the myeloma cell and the protective interaction between the tumor and its bone marrow microenvironment. Moreover, advances in our understanding of multiple myeloma pathogenesis have helped further define the intricacies of this complex disease. This book provides a concise overview of the state-of-the-art in multiple myeloma and should be of primary interest to clinicians as well as scientists and related caregivers alike in this rapidly changing f.

The objective of the current Phase II proposal is to test a novel TGF-beta based gene therapy in prostate cancer. During the Phase I study, our findings demonstrate that TGF-beta over-production by the tumor cells was immunosuppressive. In the Phase II research we proposed to render host immune cells insensitive to TGF-beta by introducing a domina negative TGF-beta type II receptor (dnTBRII) to host immune cells. Three tasks were proposed. In Task 1, we proposed to generate the dnTBRII construct with green fluoresce protein (GFP) as a selectable marker and introduce the construct into host's bone marro cells. In Task 2, we proposed to test if immune cells will escape the negative selection in the thymus. In Task 3, we proposed to test whether or not the TGF-beta insensitive immune cells will mediate anti-tumor activity.

Transforming growth factor-[beta] (TGF-[beta]), identified nearly three decades ago, is a secreted polypeptide that functions in critical cell cycle processes, including cellular proliferation, differentiation, and development: It belongs to a large protein family that, in humans, contains 33 members, including activins, inhibins, bone morphogenetic proteins, growth and differentiation factors, and Mullerian inhibiting substance. This volume draws on the world's leading laboratories to comprehensively cover all aspects of the biology of TGF-[beta] and related factors. In addition to providing historical and background information, it describes the cell biology and signaling pathways of TGF-[beta] members in detail, including the roles of TGF-[beta] factors in the development and physiology of humans and model organisms. The last few chapters are devoted to the role of TGF-[beta] members in cancer and other diseases, as well as the possibilities for therapeutics based on knowledge of signaling pathways and macromolecular structures. It serves as a comprehensive reference work for both specialists and researchers less familiar with the field.
