The induction of immune responses against tumor cells by vaccination is rapidly evolving as a therapeutic modality with new potentials for the treatment of cancer. It is based on the fact that our immune system can identify tumor cells and, once activated, is capable of developing specific immunity against the neoplastic cells. Numerous observations and intense research clearly document the major contribution of the immune system to the prevention of cancer. And there are many re ports of patients suffering from malignant melanoma or other tumors who mount a spontaneous immune response against their tumor cells that results in tumor regression. Based on the recent advances in our understanding of the compo nents of our immune system, their interactions and the regulation of immune responses, we are now able to design vaccination strategies that induce or enhance cell-mediated immunity against tumors. A ma jor advancement came with the identification and characterization of relevant tumor antigens, which are suitable target structures for anti-tu mor immune response. First clinical trials using such vaccine strategies have yielded encouraging results in patients. However, in spite of many reported cases of successful therapy of cancer by vaccination many patients still do not experience relief after such treatments. These initial clinical trials and the accompanying investigations have revealed a number of important results that indicate the direction of future re search and development in the field.

Foot-and-mouth disease (FMD) control and/or eradication efforts have been in existence for many years in different regions, supported by an official OIE system for recognition of national control programmes and of country status to manage the trade risks for FMD reintroduction. A global FMD control strategy was announced by FAO and OIE in 2012, incorporating a progressive control pathway for FMD (PCP-FMD) that elaborates principles for the application of control measures in a step-wise manner. The OIE Performance of Veterinary Services tool helps countries to monitor the structures essential for programme implementation.Vaccination is an important component of programmes that seek to reduce the impacts of FMD and to block circulation of the causative virus in order to establish and maintain disease freedom. Choosing and successfully implementing the appropriate vaccine and vaccination regimens are affected by many dynamic factors, including (1) the diversity of the viruses to be controlled; (2) the performance characteristics and instability of vaccines; (2) the range of susceptible animal species and husbandry systems; (3) the purposes of vaccination; (4) the short-lived nature of vaccine induced immunity; and (5) the design and application of vaccination programmes. Furthermore, vaccination is unlikely to succeed unless supported by other complementary control measures. Therefore, the entire process of vaccine selection and vaccination must be continuously monitored and evaluated to ensure that it fulfils its objectives and contributes to a sustainable control of FMD. This document is intended to help guide this process. Since the variable and changing circumstances of FMD control require different approaches, the guidance is not prescriptive. Instead, it reviews the options available for vaccine selection and vaccination strategies and presents methodology to check that a potential vaccine is able to provide a protective immune response and that the implemented vaccination programme has translated this into a protective level of population immunity.

Vaccination is one of the most efficient and cost effective methods of promoting human health and has been in clinical use for at least 200 years. Nevertheless, infectious diseases continue to constitute a constant threat to the well being of humanity. Common pathogens, once believed to be under control, acquire increased virulence and resistance to drugs, while exotic microorganisms emerged from hidden reservoirs to cause yet incurable diseases in humans. These changes, together with epidemic outbreaks related to political and socio-economic instabilities, increase the needs for the development of new, advanced vaccines. In this volume, devoted to the proceedings of the 39th OHOLO Conference, we present some of the recent strategies for the design and production of novel vaccines. The advent of recombinant DNA technology has stimulated the production of several subunit vaccines. In spite of the obvious advantages to this approach, the limited immuno genicity of many subunit candidates has hindered their development. Strategies to enhance the immunogenicity of subunit vaccines is therefore critical. Several approaches toward this goal, including design of novel adjuvants and delivery systems as well as design of advantageous carriers, are presented here. Among the carriers evaluated here are polypep tides (flagellin, HBV core antigen, J3-galactosidase), attenuated virions (Vaccinia, Sindbis), and nonpathogenic licensed bacteria (Salmonella).
