Learning and memory are believed to depend on plastic changes of neuronal circuits due to activity-dependent potentiation or depression of specific synapses. During the last two decades, plasticity of brain circuits was hypothesized to mainly rely on the flexibility of glutamatergic excitatory synapses, whereas inhibitory synapses were assumed relatively invariant, to ensure stable and reliable control of the neuronal network. As a consequence, while considerable efforts were made to clarify the main mechanisms underlying plasticity at excitatory synapses, the study of the cellular/molecular mechanisms of inhibitory plasticity has received much less attention. Nevertheless, an increasing body of evidence has revealed that inhibitory synapses undergo several types of plasticity at both pre- and postsynaptic levels. Given the crucial role of inhibitory interneurons in shaping network activities, such as generation of oscillations, selection of cell assemblies and signal integration, modifications of the inhibitory synaptic strength represents an extraordinary source of versatility for the fine control of brain states. This versatility also results from the rich diversity of GABAergic neurons in several brain areas, the specific role played by each inhibitory neuron subtype within a given circuit, and the heterogeneity of the properties and modulation of GABAergic synapses formed by specific interneuron classes. The molecular mechanisms underlying the potentiation or depression of inhibitory synapses are now beginning to be unraveled. At the presynaptic level, retrograde synaptic signaling was demonstrated to modulate GABA release, whereas postsynaptic forms of plasticity involve changes in the number/gating properties of GABAA receptors and/or shifts of chloride gradients. In addition, recent research indicates that GABAergic tonic inhibition can also be plastic, adding a further level of complexity to the control of the excitatory/inhibitory balance in the brain. The present Topic will focus on plasticity of GABAergic synapses, with special emphasis on the molecular mechanisms of plasticity induction and/or expression.

GABAergic neurons provide most of the inhibitory drive of adult networks and play major roles in the integrative properties of brain networks. Alterations of GABAergic signals are associated with major neurological disorders and GABA mimetic drugs are widely used as comfort molecules and to treat several brain disorders. Thus, the inhibitory drive is altered in epilepsies, infantile developmentally related disorders but also Parkinson disease, and anoxo-ischemic insults to just name a few. From the developmental stand point, GABAergic neurons mature before principal neurons and provide an important source of early activities and are thus instrumental in activity dependent modulation of the construction of cortical functional units. Gliomas are neoplasms originating in glial cells, the most malignant and frequent primary tumors of the Central Nervous System (CNS), representing approximately 40-50% of all intracranial tumors. Following the World Health Organization (WHO) guidelines, gliomas are classified into grades I-IV, regarding their malignancy degree. The latest report from WHO on CNS tumors considers that glioblastoma (GB), a grade IV adult-type diffuse glioma comprising isocitrate dehydrogenase (IDH)-wildtype tumors. Despite all efforts to improve patient survival and quality of life, the median survival remains lower due to the lack of effective diagnostic tools, poor prognosis, and limited therapeutic options. Regarding this, the design of novel treatment strategies is of the utmost importance. This can be by either the discovery of new medicines (for example synthetic compounds), repurposing known drugs or exploring novel strategies of drug delivery by nanotechnological platforms to improve treatment response and long-term survival of patients. Inter and intra-tumor heterogeneity mainly contribute to the poor prognosis and treatment efficacy. Therefore, exploiting tumor microenvironment (TME) and understanding GB’s cellular and molecular aspects are fundamental to developing more effective and better-tolerated personalized therapies. Thus, addressing physiological mechanisms involved in cancer development, progression and recurrence is crucial. Concerning this, our goal with this research topic is to share the latest advances in tumor-specific therapies for gliomas, especially GB, as well as to provide experimental validation of new bioactive molecules and their antitumor mechanism of action. Authors are invited to submit original research, review articles and systematic reviews covering, but not restricted to, the following topics: 1) Novel targeted therapies; 2) Mechanism of action and affected signaling pathways; 3) Repurposing drugs; 5) New chemical entities; 6) Pharmaceutical nanotechnology; 7) Drug delivery systems; 8) Preclinical and clinical studies of therapeutic molecules; 9) Potential molecular biomarkers to targeted therapies using proteomics or transcriptomics.
