Gene Targeting and Embryonic Stem Cells is a practical guide designed for the rapidly growing number of researchers who are moving into this field. Provides details on how to culture, transfect and differentiate established cell lines, and how to isolate new cell lines. Gene targeting experiments are described for a number of cell types, including ungulate fetal fibroblasts, murine ES cells, human embryonal carinoma cells and human ES cells, and include protocols for gene-targeting vectors, DNA transfection and RNA interference. The recent isolation of human embryonic stem cells and the potential of these cells for therapeutic applications has generated an entirely new methodology. Similarly, gene targeting methodology has recently been extended to nuclear donor cells in ungulate species. This volume will be invaluable for both new and established researchers in the field of human embryonic stem cells, and to biotech companies engaged in the production of transgenic proteins in livestock, xenotransplantation and the development of animal models.

As the major task of sequencing the human genome is near completion and full complement of human genes are catalogued, attention will be focused on the ultimate goal: to understand the normal biological functions of these genes, and how alterations lead to disease states. In this task there is a severe limitation in working with human material, but the mouse has been adopted as the favored animal model because of the available genetic resources and the highly conserved gene conservation linkage organization. In just of ten years since the first gene-targeting experiments were p- formed in embryonic stem (ES) cells and mutations transmitted through the mouse germline, more than a thousand mouse strains have been created. These achievements have been made possible by pioneering work that showed that ES cells derived from preimplantation mouse embryos could be cultured for prolonged periods without differentiation in culture, and that homologous rec- bination between targeting constructs and endogenous DNA occurred at a f- quency sufficient for recombinants to be isolated. In the next few years the mouse genome will be systematically altered, and the techniques for achi- ing manipulations are constantly being streamlined and improved.

The goal of this work is summarize the contribution that insertional mutagenesis has made to our understanding of cancer. A variety of insertional mutagens are presented that have been used to study a variety of tumor types in several model organisms. In addition, the impact of insertional mutagenesis in several gene therapy trials is discussed along with strategies to avoid such complications in future clinical trials.

An introduction for beginners and a bench reference generally to the techniques now available by which scientists can create essentially any mutation in the germline of mice by using recombination and embryonic stem cells (ESC). Genetic researchers, most from the US but also representing Britain, Canada, and Germany, cover principles and practice in mammalian cells, site-specific recombination in cells and mice, producing targeted ESC clones, producing chimeras by blastocyst and morula injection of targeted ESC, producing and analyzing ESC aggregation, gene trap strategies, and classical genetics and gene targeting. The appendix lists suppliers and contact information but not products. Annotation copyrighted by Book News, Inc., Portland, OR

The post-genomic era has brought new challenges and opportunities in all fields of the biology. In this context, several genome engineering technologies have emerged that will help deciphering genes function by as well as improve gene therapy strategies. Genomic modifications such as knock-in, knock-out, knock-down, sequence replacement or modification can today be routinely performed. However, in front of this large palette of methodologies scientists may experience difficulties to gather useful information’s scattered within the literature. This book aims to present the state of this field from basic mechanisms of site-directed modifications to their applications in a wide range of organisms such as bacteria, yeast, plants, insects, mammals. It will discuss the problems encountered when using the random integration strategy and present the recent advances made in targeted genome modification. Technologies based on Zinc Finger nucleases, Meganucleases, TALEN, CRE and FLP recombinase, C31 integrase, transposases and resolvases are fully detailed with their strengths and weaknesses. All these information’s will help students and experienced researchers to understand and choose the best technology for their own purposes.

Targeted Genome Editing Using Site Specific Nucleases written by Takashi Yamamoto and has been published by Springer this book supported file pdf, txt, epub, kindle and other format this book has been release on 2015-01-05 with Science categories.

This book serves as an introduction to targeted genome editing, beginning with the background of this rapidly developing field and methods for generation of engineered nucleases. Applications of genome editing tools are then described in detail, in iPS cells and diverse organisms such as mice, rats, marine invertebrates, fish, frogs, and plants. Tools that are mentioned include zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and CRISPR/Cas9, all of which have received much attention in recent years as breakthrough technologies. Genome editing with engineered nucleases allows us to precisely change the target genome of living cells and is a powerful way to control functional genes. It is feasible in almost all organisms ranging from bacteria to plants and animals, as well as in cultured cells such as ES and iPS cells. Various genome modifications have proven successful, including gene knockout and knock-in experiments with targeting vectors and chromosomal editing. Genome editing technologies hold great promise for the future, for example in biomedical research, clinical medicine, and generation of crops and livestock with desirable traits. A wide range of readers will find this book interesting, and with its focus on applications in a variety of organisms and cells, the book will be valuable for life scientists in all fields.
