Since the discovery of chemical neurotransmission by Otto Loewi in the 1920s, great progress has been made in our understanding of mol- ular mechanisms of neurotransmitter release. The last decade has seen an explosion of knowledge in this field. The aim of Molecular Mechanisms of Neurotransmitter Release is to provide up-to-date, in-depth coverage of essentially all major mole- lar mechanisms of neurotransmitter release. The contributors have made great efforts to write concisely but with sufficient background information, and to use figures/diagrams to present clearly key concepts or experiments. It is hoped that this book may serve as a learning tool for neuroscience students, a solid reference for neuroscientists, and a source of knowledge for people who have a general interest in neuroscience. The molecular mechanisms, which are responsible for the functional differences between the various types of neuronal synapses, have become one of the central themes of modern neurobiology. It is becoming increasingly clear that a misregulation of synaptogenesis and synaptic remodeling and dysfunctional neuronal synapses are at the heart of several human diseases, both neurological disorders and psychiatric conditions. As synapses present specialized cellular junctions between neurons and their target cells, it may not come as a surprise that neural cell adhesion molecules (CAMs) are of special importance for the genesis and the maintenance of synaptic connections. Genes encoding adhesive molecules make up a significant portion of the human genome, and neural CAMs even have been postulated to be a major factor in the evolution of the human brain. These are just some of the many reasons why we thought a book on neural CAMs and their role in establishing and maintaining neuronal synapses would be highly appropriate for summarizing our current state of knowledge. Without question, over the near future, additional adhesive proteins will join the ranks of synaptic CAMs and our knowledge, and how these molecules enable neurons and their targets to communicate effectively will grow.

The interaction of neurotransmitters, neuromodulators and neuroactive drugs with receptors localized at the cell surface initiates a chain of molecular events leading to integrated neuronal responses to the triggering stimuli. Major advancements in the characterization and isolation of recep tor molecules have answered many quest ions regarding the nature of the ele ments that determine the specificity in these interactions. At the same time, recent studies have provided evidence that delicate regulation by intracellular enzymatic systems determines the efficiency of the stimulus response coupling process, mediates the interaction between receptors, operates in feedback control mechanisms and transduces signals from the receptors to various effector sites in a highly coordinated fashion. These studies are at the focus of the present volume, which is an outcome of a symposium held at the University of Vermont College of Medicine on March 21-23, 1986, in conjunction with the seventeenth annual meeting of the Amer ican Society for Neurochemistry. The symposium has demonstrated clearly that the concerted efforts of investigators in neurophysiology, biochemis try, pharmacology, cell-biology, molecular genetics, neurology, and psy chiatry are required to achieve better understanding of the processes under lying neuronal responsiveness. Keeping abreast of the enormous advances in neuroscience in the five years since the first edition was published, the authors have revised all their chapters in the second edition. What was formerly the first chapter has been expanded substantially and divided into two separate chapters to emphasize the cell biology of neurons and glia, and their commonalities with other kinds of cells. The section on intracellular communication has also been expanded and reorganized. Levitan and Kaczmarek introduce the concept of ion channels as specialized membrane proteins at an early stage, making the idea of selective membrane permeability more accessible in terms of the properties of specific ion channel proteins. In addition, they emphasize the astonishing diversity of voltage-dependent ion channels that has become evident in recent years, and discuss the implications of this diversity for neuronal physiology. In the section on intercellular communication, the chapter on neurotransmitter secretion has also been rewritten to reflect the new level of understanding of secretion that has resulted from the identification of many of the molecular players in vesicle fusion and exocytosis. The other chapters in this section have also been fully revised to incorporate new information resulting from the cloning and characterization of the multitude of glutamate receptors as well as to describe novel elements of intracellular signaling pathways in neurons and other cells. The last decades have witnessed a radical change in our views on central nervous system damage and repair. This change is not only due to the emergence of new powerful tools for the analysis of the brain and its reactions to insults, but it also reflects a conceptual change in the way we approach these problems. As an illustration to this development, it is instructive to go back to the proceedings of a meeting at the NIH in 1955 edited by William F. Windle, which summarizes the disillusioned and pessimistic view on CNS regeneration prevailing at the time. While this generation of researchers were well aware of the issues at stake, they felt they had reached the end of the road; the approaches they had pursued had got stuck and the tools available could not take them any further. I can very well imagine that the participants, most of them leaders in the field, left that conference feeling they had heard their field being sentenced to death.
