Long non-coding RNAs (lncRNAs) are defined as transcripts longer than 200 nucleotides rarely translatable into protein, which distinguishes them from small non-coding RNAs (sncRNAs) such as miRNAs, siRNAs, piRNAs, snoRNAs exRNAs, (scaRNAs). Long intervening/intergenic noncoding RNAs (lincRNAs) refer to lncRNA non-overlapped to protein-coding genes. In terms of abundance and specificity, ~30,000 lncRNAs have been identified in human tissues with ~ 10- fold lower abundance than mRNA. Near 80% of lncRNAs show tissue-specific features, in contrast to only less than 20% of mRNAs. In addition to tissue specificity, lncRNAs are also characterized by having significantly higher developmental stage specificity. Of the identified lncRNAs, although only a very small proportion have been validated to be biologically relevant, the emerging evidence has confirmed important regulatory functions at levels of transcription, post transcription, and epigenetic control. Physiologically, lncRNAs are involved in growth, development, reproduction, aging, and pathogenesis of disease initiation and progression, such as neurological disorders and cancers.

Metabolic syndrome (MetS) is a set of co-morbidities that collectively increase an individual’s risk of developing cardiovascular disease, stroke, and type 2 diabetes mellitus (T2D). Per the World Health Organization (WHO), MetS is typically characterized by obesity, insulin resistance, hypertension, and hyperlipidemia. Building on this point, some of the major risk factors for development of MetS include increased weight or an obese phenotype, lack of physical activity, and genetics. Interestingly, the last decade has witnessed a deluge of Genome-Wide Association Studies (GWAS) that have linked hundreds of genomic with both collective MetS traits, as well as individual metabolic disorders sitting within it. Currently, in the post-GWAS era, the focus has shifted to characterization of these novel genomic to establish causality and disease relevance. This is being pursued by way of functional validation such as gain- and loss-of-function studies, investigating resulting metabolic phenotypes, mechanisms and pathways underlying these phenotypes, their prevalence and potential for risk stratification across populations, and finally, identification of therapeutic targets for pharmacological intervention.

Metabolic syndrome is a dangerous combination of cardiovascular risk factors that correlate with each other and can increase cardio and cerebrovascular events. The incidence of metabolic syndrome often parallels that of obesity and type 2 diabetes. It is widespread; in the United States, more than 40% of people over the age of 50 have metabolic syndrome. The diagnosis of metabolic syndrome is based on the coexistence of at least three risk factors including waist circumference greater than 102 cm in men or 88 cm in women, systolic blood pressure greater than 130 mmHg and diastolic greater than 85 mmHg, HDL cholesterol less than 40 mg/dl in men or 50 mg/dl in women, triglyceridemia higher than 150 mg/dl and finally fasting glycaemia higher than 110 mg/dl. The causes are to be found in a combination of lifestyle habits (diet and poor physical activity) and family predisposition (genetics). These causes lead to the accumulation of abdominal fat, which in turn plays a decisive role in favoring the combination of the various risk factors. Among these, insulin resistance is fundamental in the onset not only of hyperglycemia and diabetes, but also of dyslipidemia and hypertension. Metabolic syndrome does not manifest itself with specific symptoms, which is why attention should be paid to risk factors. With this in mind, we welcome researchers investigating this contemporary theme. This Research Topic welcomes original research articles, perspectives, and review articles.
