Salmonella infection, also known as Salmonellosis, is a common zoonotic food-borne infection in humans, poultry and mammals. The disease is commonly spread in humans through contaminated foods and the infection can develop into gastroenteritis, enteric fever, bacteraemia or focal disease. The genus Salmonella contains only two species, Salmonella enterica and Salmonella bongoriwhich further subdivides into a number of serotypes resulting in either typhoid Salmonella and non-typhoid Salmonella (NTS) infections. The serotypes of S. enterica such as Salmonella typhi and paratyphi are responsible for the typhoidal infections leading to enteric fever and other serotypes such as Salmonella typhimurium and enteridis are responsible for non-typhoidal infections leading to gastroenteritis and focal disease. Salmonella infections are one of the leading foods borne infections across the world resulting in socio-economic and hospital burden. There are continuous and ongoing efforts in understanding the structural features and mode of action of these pathogens vital in driving scientific discovery towards prevention and treatment strategies against these infections. Through this research topic, we aim to explore the scientific gaps associated with our understanding of Salmonella spp. and its importance to design better drug and treatment modalities.

Many of the most prevalent and devastating human and animal pathogens have part of their lifecycle out-with the animal host. These pathogens have a remarkably wide capacity to adapt to a range of quite different environments: physical, chemical and biological, which is part of the key to their success. Many of the well-known pathogens that are able to jump between hosts in different biological kingdoms are transmitted through the faecal-oral and direct transmission pathways, and as such have become important food-borne pathogens. Some high-profile examples include fresh produce-associated outbreaks of Escherichia coli O157:H7 and Salmonella enterica. Other pathogens may be transmitted via direct contact or aerosols are include important zoonotic pathogens. It is possible to make a broad division between those pathogens that are passively transmitted via vectors and need the animal host for replication (e.g. virus and parasites), and those that are able to actively interact with alternative hosts, where they can proliferate (e.g. the enteric bacteria). This research topic will focus on plants as alternative hosts for human pathogens, and the role of plants in their transmission back to humans. The area is particularly exciting because it opens up new aspects to the biology of some microbes already considered to be very well characterised. One aspect of cross-kingdom host colonisation is in the comparison between the hosts and how the microbes are able to use both common and specific adaptations for each situation. The area is still in relative infancy and there are far more questions than answers at present. We aim to address important questions underlying the interactions for both the microbe and plant host in this research topic.

The human body is constantly faced with microorganisms. Most of these bacteria, fungi, and viruses are harmless, many of them are beneficial, and a small fraction is pathogenic. For humans, infection with pathogenic microorganisms can be very serious or even fatal, ranging from mild transient or chronic infections to death. The first line of defence against pathogens is our innate immune system. Beside chemical and physical defence mechanisms of the innate immune system, phagocytic cells such as macrophages play a crucial role in the fight against pathogenic microorganisms. However, phagocytic cells and pathogens are in a constant evolutionary arms race, inventing new strategies to successfully kill pathogens and learning how to resist phagocytosis and intracellular killing, respectively. If pathogens are not obligatory adapted to the human body or other animals, they also have to face environmental phagocytes in the form of amoebae. Many aspects of phagocytosis and intracellular killing are surprisingly well conserved between amoebae and macrophages. Therefore, pathogens that have evolved with environmental amoebae as their “training grounds” can also be successful during infection of macrophages and other animal phagocytic cells. In this Research Topic, we provide the latest knowledge about the potential of using amoebae as host models to study the interaction with pathogens. The Research Topic covers the interaction of amoebae with bacteria, fungi, and viruses and also illustrates the similarities and differences between amoebae and macrophages. Investigation of evolutionary conserved pathways of amoebae and macrophages furthers our understanding of the biology of host-pathogen interactions and helps to develop new anti-infection therapies.

The use of model antigens such as haptens and ovalbumin has provided enormous insights into how immune responses develop, particularly to vaccine antigens. Furthermore, these studies are overwhelmingly performed in animals housed in clean facilities and are not known to have experienced overt clinical signs caused by infectious agents. Therefore, this is unlikely to reflect the impact more complex host-pathogen interactions can have on the host, nor the diversity in how immunity is regulated. Humans develop immune responses in the context of the periodic exposure to multiple pathogens and vaccines over a life-time. These are likely to have a long-lasting effect on who and what we are and how we respond to further antigen challenge. Therefore, studies on how infection influences immune homeostasis and how the development of responses to a pathogen reflects what is known on immune regulation will be informative on how we can translate findings from our standard models into treatments usable in humans.
One organism allows us to do just this. Bacteria of the genus Salmonella are devastating human pathogens. Nevertheless, many aspects of the diseases they cause can be successfully modelled in murine systems so that the infection is either resolving or non-resolving. This has the advantage of allowing the long-term impact of infection on immune function to be assessed. We propose to welcome key workers to write about their research that examine the consequence of Salmonella infection on the host and the elements of the bacterium that contribute to this.
