Neurodegenerative diseases represent a very large group of heterogeneous disorders affecting specific subtypes of neurons in the brain. This book contributes insight both to the awareness of the brain and its neurodegenerative states. The chapters present current knowledge regarding genetics, molecular mechanisms, and new therapeutic strategies against neurodegenerative disorders. The book is intended to serve as a source to aid clinicians and researchers in the field, and also life science readers to increase their understanding and awareness of the clinical correlations, genetic aspects, neuropathological findings, and current therapeutic interventions in neurodegenerative diseases. I believe that this book will enlighten the curiosity for neurodegeneration and also encourage researchers to work on potentially effective molecular therapies for still mysterious neurodegenerative disorders.

Causation is an aspect of epilepsy neglected in the scientific literature and in the conceptualization of epilepsy at a clinical and experimental level. It was to remedy this deficiency that this book was conceived. The book opens with a draft etiological classification that goes some way to filling the nosological void. The book is divided into four etiological categories: idiopathic, symptomatic, cryptogenic, and provoked epilepsies. Each chapter considers topics in a consistent fashion, dealing with the phenomenon of epilepsy in each etiology, including its epidemiology, clinical features and prognosis, and any specific aspects of treatment. The book is a comprehensive reference work, a catalogue of all important causes of epilepsy, and a clinical tool for all clinicians dealing with patients who have epilepsy. It is aimed at epileptologists and neurologists and provides a distillation of knowledge in a form that is helpful in the clinical setting.

Homeostatic Control Of Brain Function written by Detlev Boison and has been published by Oxford University Press this book supported file pdf, txt, epub, kindle and other format this book has been release on 2016 with Medical categories.

Homeostatic Control of Brain Function offers a broad view of brain health and diverse perspectives for potential treatments, targeting key areas such as mitochondria, the immune system, epigenetic changes, and regulatory molecules such as ions, neuropeptides, and neuromodulators. Loss of homeostasis becomes expressed as a diverse array of neurological disorders. Each disorder has multiple comorbidities - with some crossing over several conditions - and often disease-specific treatments remain elusive. When current pharmacological therapies result in ineffective and inadequate outcomes, therapies to restore and maintain homeostatic functions can help improve brain health, no matter the diagnosis. Employing homeostatic therapies may lead to future cures or treatments that address multiple comorbidities. In an age where brain diseases such as Alzheimer's or Parkinson's are ever present, the incorporation of homeostatic techniques could successfully promote better overall brain health. Key Features include · A focus on the homeostatic controls that significantly depend on the way one lives, eats, and drinks. · Highlights from emerging research in non-pharmaceutical therapies including botanical medications, meditation, diet, and exercise. · Incorporation of homeostatic therapies into existing basic and clinical research paradigms. · Extensive scientific basic and clinical research ranging from molecules to disorders. · Emerging practical information for improving homeostasis. · Examples of homeostatic therapies in preventing and delaying dysfunction. Both editors, Detlev Boison and Susan Masino, bring their unique expertise in homeostatic research to the overall scope of this work. This book is accessible to all with an interest in brain health; scientist, clinician, student, and lay reader alike.

Although more than 10 new antiepileptic drugs have been developed in the past decade, epilepsy remains resistant to drug therapy in about one third of patients, many of whom struggle with the disease their entire lives. Managing these patients is a challenge and requires a structured multidisciplinary approach. The book includes chapters on all issues related to pharmacoresistance in epilepsy and describes recent developments in the pathogenesis and treatment of this disorder. It addresses abnormalities in inhibitory mechanisms, epilepsy-related changes to the immune system, development of pharmacoresistance caused by chronic exposure to antiepileptic drugs, and novel therapeutic strategies for preventing or slowing down the progression of the disease. Clinicians and basic scientists alike will find up-to-date information on the development of pharmacoesistance, as well as reviews of mechanisms associated with epilepsy that may help them consider novel strategies for preventing the development of pharmacoresistance in the first place. The book also features information on new therapeutic strategies for control of epilepsy, such as transcutaneous electrical stimulation and virtual screening of new antiepileptic drugs. Pharmacoresistance in Epilepsy: From Genes and Molecules to Promising Therapies is useful to anyone working in the field, whether they’re studying epilepsy in the lab or treating it in a doctor’s office.”

Epilepsy is one of the most common neurological chronic disorders characterized by unpredictable recurrent seizures, which leads to brain dysfunction, cognitive, psychological, and social disorders. Various treatment options for epilepsy are: antiepileptic drugs, surgery, neuromodulation, gene therapy and ketogenic diet. However, definitions, diagnosis and treatment of epilepsy are still challenging issues for clinicians and researchers. Antiepileptic drugs play a central and crucial role in the treatment of epilepsy patients. Brivaracetam Cannabidiol (CBD), Cenobamate, Everolimus and Fenfluramine are the five novel anticonvulsant drugs during the last 5 years. Brivaracetam anticonvulsant effects are due to its highly selective affinity for synaptic vesicle protein 2A (SV2A) in the brain. It is act as an SV2A ligand. Among these drugs Cannabidiol is one of the prominent phytocannabinoids derived from the cannabis sativa. The anticonvulsant mechanisms of action of CBD including antagonism of G protein-coupled receptor 55 (GPR55), desensitization of transient receptor potential vanilloid type 1 (TRPV1) channels, and potentiation of adenosine-mediated signaling through inhibition of the equilibrative nucleoside transporter 1 (ENT-1) and enhancement of GABAergic transmission. The mechanisms of the antiseizure effect of Cenobamate are: increasing the inactivated state of voltage-gated sodium channels, and acting as a positive allosteric modulator of the GABA-A receptors by binding at a non-benzodiazepine site. Everolimus is a mammalian target of rapamycin (mTOR) inhibitor. Fenfluramine is able to enhance extracellular serotonin levels, modulate serotonergic and other neurologic receptors. Nevertheless, the exact mechanism of action of most of these drugs is unknown. At least 30% of people with epilepsy have drug-resistant epilepsy (DRE), therefore, non-pharmacological treatments have been considered. the Vagus Nerve Stimulation (VNS), Trigeminal Nerve Stimulation (TNS), Transcranial Direct Current Stimulation (tDCS), Transcranial Magnetic Stimulation (TMS) are the most common neuromodulation-based approaches. Also, in recent years, a new field of GABA-, adenosine-, galanin-, and neuropeptide Y- based cell and gene therapies for refractory epilepsy has emerged. In spite of these advances, significant gaps in knowledge still remain. In particular, the mechanisms underlying epilepsy, including status epilepticus, drug and surgical treatments are still not well elucidated. Novel insights on pathophysiological mechanisms of epilepsy and new treatment options prompted us to collect the most recent publications on pharmacological and non-pharmacological treatments of epilepsy. So, in this Research Topic, we will focus on the advances in the field of treatment in epilepsy, the pathophysiology and molecular aspects of seizure attacks and novel antiseizure medications.
