Tumors and their surroundings are closely related and constantly interact. Tumors can influence their microenvironment by releasing cell signaling molecules that promote neoplasm angiogenesis and induce immune tolerance, while immune cells in the microenvironment can influence cancer cell growth and development. The tumor microenvironment (TME), a complex biological ecosystem for cancer cells to survive and develop refers to the surrounding circumstances of cancer cells, including surrounding blood vessels, immune cells, fibroblasts, bone marrow inflammatory cells, various signaling molecules and extracellular matrix (ECM). The immune cells and their regulation mode in TME have tumor-antagonizing or tumor-promoting functions. TME has been gradually recognized as a key contributor to cancer progression and drug resistance, with cellular components in the TME able to enhance tumor resistance by recruiting and secreting multiple protective cytokines. The acellular components of TME can mediate drug resistance by building physical barriers, affecting tumor cell growth and metabolism, etc. Knowledge of the above is paving the way for identifying new targets and discovering new therapies. Studying the dynamic relationship between tumor surroundings and neoplasm, and clarifying the molecular mechanism of different factors refers to TME in the process of tumor progression, are the key elements of cancer inhibition. Currently, there is a proliferation of publicly available tumor genomic databases. Such as the Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), Surveillance, Epidemiology, and EndResults Program (SEER), International Cancer Genome Consortium (ICGC) and National Cancer Database (NCDB). These abundant public database resources enable researchers to mine multi-omics cancer data to better understand the interactions between tumors and their microenvironment and to discover biomarkers and therapeutic targets in TME that are associated with tumorigenesis. This provides new targets for early diagnosis and precise treatment of tumors. In this research topic, we would like to demonstrate the function and mechanism of tumor microenvironment in tumorigenesis and development through various methods, including sequencing, bioassay, experimental models such as PDX and organoids. This Research Topic is the second volume of the “Novel Biomarkers in Tumor Immunity and Immunotherapy” Community Series. Please see Volume I here. Immune checkpoint inhibitors (ICIs) such as anti-PD-1 and anti-PD-L1 antibodies are highly effective against many types of cancer, yet durable responses are limited to a subset of patients highlighting the need for the development of effective biomarkers to predict prognosis and efficacy. Currently, PD-L1 expression in tumors, microsatellite instability (MSI) or mismatch repair deficiency (dMMR), and tumor mutation burden (TMB) are known as biomarkers for cancer immunotherapy but are not sufficient. Combination therapy with immune checkpoint inhibitors and chemotherapy or radiation therapy, as well as diverse therapies targeting intra-tumoral regulatory T cells have been described, but there are currently no unifying biomarkers that are applicable to clinically, a simple, fast, non-invasive method that can yield biomarkers of disease with a minimal adverse effect on patients is desirable. Recent findings suggest that the balancing of effector T cells and regulatory cells in the tumor microenvironment is associated with cancer progression and prognosis. Cells and molecules involved in the control of cancer are complex, and a better understanding of the tumor immune environment will lead to the development of truly effective biomarkers. This research topic will focus on novel biomarkers that predict efficacy, prognosis, or the development of adverse events in various cancer immunotherapies, and extensive basic research leading to the development of biomarkers. We expect a wide range of research, not only in serology, genetics, and immunocytochemistry but also in bacterial flora. Research on the development of novel assays and bioinformatics methods is also welcome: • Non-invasive biomarkers for cancer immunotherapy. • Bulk RNA-seq, scRNA-seq, or Rep-seq methods. • Correlation of tumor immune cells with gut microbiota in tumor immunotherapy. • Impact of Teff and Treg balance in the tumor microenvironment on tumor prognosis. • Inflammatory and immune signatures associated with drug response versus resistance in cancer. The tumor microenvironment (TME) plays a critical role in tumor proliferation, progression, and therapeutic responses. TME is a complex network of cancer cells, stromal cells, and, most importantly, infiltrating immune cells. Cancer cells regulate numerous biological functions through direct or indirect interaction with TME components. Emerging evidence suggests that TME crucially influences the response to both chemotherapy and immunotherapy. As scientific research has entered the big data era with the fast development of high-throughput sequencing technologies, machine learning has been gradually widely applied to extract important knowledge from big data bioinformatics.
