In general, infection of a host by multiple pathogens has important clinical implications. In conjunction with HIV, co-infections are associated with a significantly decreased survival probability. Unexpectedly, long-term viremia of the apathogenic GB Virus C (GBV-C) leads to improved survival of GBV-C co-infected HIV patients. However, epidemiological studies cannot prove a causal relationship nor they distinguish whether the observed reduction in HIV associated morbidity and mortality is caused by GBV-C/HIV interference or if GBV-C serves just as a surrogate marker for a robust immune system. Therefore, cell culture experiments were necessary. Indeed, GBV-C inhibits different HIV strains and clades. The underlaying mechanism of HIV suppression is complex. In addition to GBV-C proteins, such as the E2 glycoprotein that possess the ability to inhibit HIV entry or the NS5A protein that may imair later HIV replication steps, GBV-C elicits antibodies that neutralize HIV comparable to the broad reactive human anti-gp41 antibodies 2F5 and 4E10, as well. Taken together a combination of both inhibitory effects present an innovative approach for further drug development and vaccine design.

Interferons (IFNs) play pivotal roles in shaping the immune responses in mammals and are particularly important for the control of viral infections and cell growth, and immune regulation. These proteins rapidly induce an 'anti-viral state' in cells that surround infected cells. In order to survive, viruses have evolved multiple strategies to evade the anti-viral effects of IFNs. Elucidating the molecular and cellular biology of the virus-interferon interaction is key to understanding issues such as viral pathogenesis, latency, and the development of novel antivirals. In this book, a panel of international experts reviews the current hot-topics, producing a timely overview of this exciting field. The book opens with a chapter that comprehensively reviews the antiviral effects of extracellular double-stranded RNA, the 'viral toxin'. This is followed by chapters that review the properties of type I and type III interferons, and the role of interferon-stimulated genes. The next five chapters are devoted to understanding the diverse strategies used by clinically relevant human viruses to subvert host interferon responses. The closing chapter provides an interesting overview of the clinical application of interferon as antiviral and anticancer agents. Essential reading for every scientist involved in interferon or antiviral research and a recommended text for all virology laboratories.

The CRISPR/Cas9 system provides bacteria and archaea with molecular immunity against invading phages and conjugative plasmids. Recently, CRISPR/Cas9 has been used for targeted genome editing in diverse eukaryotic species. These data establish the efficacy of the CRISPR/Cas9 system for viral interference in plants, thereby extending the utility of this technology and opening the possibility of producing plants resistant to multiple viral infections. Proceeds from the sale of this book go to the support of an elderly disabled person.
