Naïve T cells get activated upon encounter with their cognate antigen and differentiate into a specific subset of effector cells. These T cells are themselves plastic and are able to re-differentiate into another subset, changing both phenotype and function. Differentiation into a specific subset depends on the nature of the antigen and of the environmental milieu. Notably, certain nutrients, such as vitamins A and D, sodium chloride, have been shown to modulate T cell responses and influence T cell differentiation. Parasite infection can also skew Th differentiation. Similarly, the gut microbiota regulates the development of immune responses. Lastly, the key role of metabolism on T cells has also been demonstrated. This series of articles highlights some of the multiple links existing between environmental factors and T cell responses.

The development, clinical translation and recent efficacy of novel gene therapies targeting refractory malignancies has led to research that extends this technology to a variety of infectious and rheumatological diseases. Unlike conventional drugs or antibodies, T cells have the potential to target and exert effector function in response to disease in a dynamic manner, acting as a “living drug”. The most efficacious form of gene-modified T cells to date is the chimeric antigen receptor (CAR)-modified T cell, which redirects the specificity of T cells to an antigen expressed by tumor cells. Clinical experience with autologous CAR-T cells, primarily in hematologic malignancies, has underscored the feasibility and safety of the approach, while also demonstrating dramatic and sustained antitumor effects through mechanisms orthogonal to those of traditional anticancer therapies. However, several challenging obstacles must be surmounted in order to improve the broader efficacy of this approach.

Lymphocytes constantly survey the lymph nodes in search for potential infection by a pathogen. They enter the afferent lymphatic vessel that serves as a conduit to transport the motile lymphocytes to the draining lymph node. Lymphatic vessels (LVs) are present in most vascularized tissues. They are traditionally regarded as passive conduits for soluble antigens and leukocytes. Afferent LVs begin as blind ended capillaries, which give rise to collecting vessels that merge and connect with draining lymph nodes (dLNs). Initial lymphatic capillaries are composed of Lymphatic Endothelial Cells (LECs) connected by discontinuous cell junctions, which join to form larger collecting lymphatic vessels, and ultimately feed into the LN subcapsular sinus. Within the LN, LECs are localized to the subcapsular, cortical, and medullary sinuses, where they interact with incoming and exiting leukocytes. LECs, and in general LN stromal cells, have emerged in the recent years as active players in the immune response. In support to this,studies have shown that the immune response generated during inflammation and under pathologic conditions is accompanied by modeling of the LVs and generation of new lymphatics, a process known as lymphangiogenesis. These facts strongly suggest that LECs and stromal LN cells in general, are not inert players but rather are part of the immune response by organizing immune cells movement, exchanging information and supplying survival factors. The purpose of this research topic is to review the role of the LECs during immune homeostasis and cancer. Considering the critical role of lymphangiogenesis in many pathologies like chronic and acute inflammation, autoimmunity, wound healing, graft rejection, and tumor metastasis, it is important to understand the molecular mechanisms that govern the cross talks between the LECs and immune cells during homeostasis and inflammation.

Long-lasting T cell immunity is delivered by an array of individual T lymphocytes expressing clonally distributed and highly specific antigen receptors recognizing an almost infinite number of antigens that might enter in contact with the host. Following antigen-specific priming in lymphnodes, naïve CD4 and CD8 T lymphocytes proliferate generating clones of effector cells that migrate to peripheral tissues and deliver unique antigen-specific effector functions. Moreover, a proportion of these effector lymphocytes survive as memory T cells that can be rapidly mobilized upon new exposure to the same antigen, even years after their primary induction. Innate immune cells play crucial roles in the induction and maintenance of this efficient protection system. Following the seminal discovery of Steinman and Cohen in 1974 describing a rare cell type capable of initiating antigen-specific responses in lymphnodes, Dendritic Cells (DC) have taken up the stage for several decades as professional Antigen Presenting Cells (APC). Although DC possess all attributes to prime naïve T lymphocytes, other immune cell subsets become crucial accessory cells during secondary and even primary activation. For instance, Monocytes (Mo) are rapidly recruited to inflammatory sites and have recently been recognized as capable of shaping T cell immunity, either directly through Ag presentation, or indirectly through the secretion of soluble factors. In addition, upon sensing of T cell-derived cytokines, Mo differentiate into functionally different APC types that further impact on the quality and persistence of memory T cell responses in peripheral tissues. Other innate immune cells, including Myeloid Derived Suppressor Cells, Granulocytes and iNKT lymphocytes, are known to modulate T cell activation by interacting with and modifying the function of professional APC. Notably, innate immune cell determinants also account for the tissue-specific regulation of T cell immunity. Hence, the newly discovered family of Innate Lymphoid Cells, has been recognized to shape CD4+ T cell responses at mucosal surfaces. Although the actions of innate immune cells fulfills the need of initiating and maintaining protective T cell responses, the excessive presence or activity of individual determinants may be detrimental to the host, because it could promote tissue destruction as in autoimmunity and allergy, or conversely, prevent the induction of immune responses against malignant tissues, and even modulate the response to therapeutic agents. Thus, understanding how defined innate immune cell subsets control T cell immunity is of fundamental relevance to understand human health, and of practical relevance for preventing and curing human diseases. In this research topic, we intend to provide an excellent platform for the collection of manuscripts addressing in depth how diverse innate immune cell subsets impact on T cell responses through molecularly defined pathways and evaluating the rational translation of basic research into clinical applications.
